More than ten percent of people suffer from at least one episode of depression and related mental disorders in a lifetime, and depression and related mental disorders are one of the world’s greatest public health problems. A multiple system theory holds that dysregulation of the multiple systems underlies the pathogenesis of depression and related mental disorders, and new therapies based on the multiple system dysregulation theory are urgently needed. In this study, the antidepressant effect of decoction from herbi Cistanche deserticola/i Y.C.Ma andi Cistanche tubulosa/i was examined. Herbi Cistanche/i decoction reduced the immobility period significantly in the mouse tail suspension test. Mice treated with herb decoction showed an improved ability of spatial learning and memory in the Morris water maze test. Groups treated herb decoction displayed a downregulated monoamine oxidase (MAO) activity; the dopamine (DA) concentration in the brain was upregulated, indicating herbi Cistanche/i decoction improved the nerve excitability; the serum concentration of corticosterone (CORT) was downregulated, showing that mice benefited from a reduced stress level. Hence, the antidepressant efficacy and mechanism of traditional Chinese herbi Cistanche/i were explored in this study. Herbi Cistanche/i showed a potential to be developed as a complementary and alternative therapy for depression.

Epigenetic “clocks” based on DNA methylation (DNAme) are the most robust and widely employed aging biomarker. They have been built for numerous species and reflect gold-standard interventions that extend lifespan. However, conventional methods for measuring epigenetic clocks are expensive and low-throughput. Here, we describe Tagmentation-based Indexing for Methylation Sequencing (TIME-Seq) for ultra-cheap and scalable targeted methylation sequencing of epigenetic clocks and other DNAme biomarkers. Using TIME-Seq, we built and validated inexpensive epigenetic clocks based on genomic and ribosomal DNAme in hundreds of mice and human samples. We also discover it is possible to accurately predict age from extremely low-cost shallow sequencing (e.g., 10,000 reads) of TIME-Seq libraries using scAge , a probabilistic age-prediction algorithm originally applied to single cells. Together, these methods reduce the cost of DNAme biomarker analysis by more than two orders of magnitude, thereby expanding and democratizing their use in aging research, clinical trials, and disease diagnosis. ### Competing Interest Statement P.T.G. and D.A.S. are named inventors on a patent application related to TIME-Seq methods filed by Harvard Medical School and licensed to Longevity Sciences. t and hoarseness. METHODS: One hundred fifty patients, ASA physical status I to II, scheduled to undergo general anesthesia and elective orthopedic or gynecologic surgery were enrolled. Participants were randomly allocated to receive either Strepsils or identical-looking placebo tablets immediately before arrival to the operating room. The incidence and severity of postoperative sore throat and hoarseness were evaluated immediately and 24 hours after surgery. RESULTS: The incidence of early postoperative sore throat was 13.7% and 33.3% and hoarseness was 12.3% and 26.4% in the Strepsils and placebo groups, respectively (P 0.05). One day after surgery, the incidence of sore throat decreased to 6.8% and 18.1% in the Strepsils and control groups, respectively. The incidence of hoarseness 1 day after the operation decreased to 8.2% in the Strepsils group and 19.4% in the placebo group, but the difference remained statistically significant (P 0.05). CONCLUSION: Perioperative use of Strepsils tablets reduces postoperative sore throat and hoarseness of voice....
