Cowboys Vs. Eagles Live Score, Updates, Highlights From NFL ‘Sunday Night Football’ Game – Sporting News https://dentoncountynewsonline.com/cowboys-vs-eagles-live-score-updates-highlights-from-nfl-sunday-night-football-game-sporting-news/ Two of the NFL’s best teams will face off under the lights in an early-season divisional showdown on “Sunday Night Football.” Oh, yeah, they also don’t like each other very much. The Cowboys and Eagles meet for the first time this season, in prime time in Week 6 — and the stakes couldn’t be much higher. Both teams have gotten off to outstanding starts in 2022. Philadelphia is the only undefeated team remaining in the league at 5-0 while Dallas is lurking at 4-1. Eagles quarterback Jalen Hurts has forced his way into the MVP discussion with 10 total touchdowns (four passing, six rushing). His supporting cast on offense — including A.J. Brown, DeVonta Smith and Miles Sanders — is making life difficult for opposing defenses. That said, Philly has only played one team with a winning record thus far (the Vikings in Week 2). This week’s game will be one of its toughest tests to date. MORE: See where Cowboys, Eagles stack up in latest NFL power rankings Cowboys fans could be forgiven for thinking their season was over after a Week 1 loss to the Buccaneers in which Dak Prescott suffered a thumb fracture on his throwing hand. But they’ve rattled off four wins in a row since then with Cooper Rush filling in at quarterback. The hallmark of this team is its defense, with reigning DROY Micah Parsons spearheading a pass rush that’s second in the league in sacks per game. The unit is also allowing just 14 points per game, which was good for third in the NFL after five weeks. Rush has been winning games but he hasn’t necessarily been lighting it up. He’s coming off his worst statistical game of the season: 10-for-16 passing, 102 yards, no touchdowns (but also no interceptions) in a win over the Rams. He’ll need to play better than that to keep up with an explosive Eagles offense. In the ad, via Colossus, Banks puts on a pair of sunglasses and decides that she’s in the metaverse. “I do look skinnier,” Banks says, patting her stomach. She then runs into Marian Leitner-Waldman, who runs the company with her husband, David Waldman. “What are you doing?” Leitner-Waldman asks. “You look so real,” says Banks. “I’m in the metaverse.” Leitner-Waldman then tells Banks she’s wearing her sunglasses. Banks shushes Leitner-Waldman and gives her a “non-fungible token” (a vase) and Leitner-Waldman walks off. advertisement advertisement

2007 IPhone Sells For $53,000 At Auction – Gizmodo Australia https://dentoncountynewsonline.com/2007-iphone-sells-for-53000-at-auction-gizmodo-australia/ In the market for a new iPhone? Unfortunately, you might have to settle for one of Apple’s 2022 models, after an auction house in California just let a 2007 iPhone go for a cool $US32,783 – around $53,000 in Aussie terms. LGC Auctions was flogging an original 2007 iPhone that is still factory sealed. The box looks more like a Cards Against Humanity expansion pack than it does an iPhone box. Bidding for the iPhone started at $US2,500 (~$4,000) and at about 10:15 AEDT on Monday, some sucker (yes, I’m jealous) had committed $US32,783 to the cause. When it was first launched in 2007, the iPhone retailed for around $US600. The auction lot description describes the phone as being “one of the most important and ubiquitous inventions of our lifetime” and while we definitely agree, a home deposit (almost) seems like ridiculous money to drop on a gadget that’s all-but a cool paperweight. The 2007 iPhone auctioned off boasts a whopping 8GB of storage, a touchscreen, a 2MP camera (lol) and, get this, a web browser. Honestly this is a fantastic find and it’s great looking back at how much things have changed. For comparison, the iPhone 14 Pro Max has a 48MP camera and will come with 1TB of storage if you want to hand over $2,769. This factory sealed, first-release iPhone is in exceptional condition. As per the pictures, and testimony from the auction house, the packaging is virtually flawless along the surface and edges, the factory seal is clean with correct seam details and tightness and labels on the reverse are correctly pristine beneath the seal. Image: LGC Auctions “Collectors and investors would be hard-pressed to find a superior example.  Relevance and rarity comprise a winning formula for this red hot collectible,” they wrote. Here’s Steve Jobs announcing the iPhone back in 2007 if you’re after some more nostalgia.

CFTC Action Shows Why Crypto Developers Should Get Ready To Leave The US Cointelegraph
CFTC Action Shows Why Crypto Developers Should Get Ready To Leave The US Cointelegraph
CFTC Action Shows Why Crypto Developers Should Get Ready To Leave The US – Cointelegraph https://dentoncountynewsonline.com/cftc-action-shows-why-crypto-developers-should-get-ready-to-leave-the-us-cointelegraph/ Considerable anxiety exists in the world of Web3 related to regulation and the legal status of cryptocurrency projects. It’s particularly apparent in the United States, where the Commodity Futures Trading Commission (CFTC) fueled concerns in September with an announcement that it was imposing a $250,000 fine on a decentralized autonomous organization (DAO), Ooki DAO, and its investors. The fine was particularly ominous, considering DAOs are intended to be “regulation proof.” The CFTC said in its statement on the issue that Ooki DAO’s bZeroX protocol offered illegal off-exchange trading of digital assets. The agency took issue with the fact that the founders, Tom Bean and Kyle Kistner, tried to use the existing bZeroX protocol within the DAO to put it beyond the reach of regulators. “By transferring control to a DAO, bZeroX’s founders touted to bZeroX community members the operations would be enforcement-proof,” the CFTC said. “The bZx Founders were wrong, however. DAOs are not immune from enforcement and may not violate the law with impunity.” The fine is not all that surprising. The CFTC and other regulators are not going to abide by a veil of decentralization. But, there is something within the ruling that is extremely worrying to Web3 lawyers and developers. The agency’s complaint indicated that the voters within a given DAO could be distinctly liable. In other words, no longer will only founders be targeted, as users who take part could also be liable. This is sure to have a chilling effect on turning people away from DAOs and Web3 in general. After all, the whole point is to avoid this kind of targeting and to create new ecosystems where all parties can vote in peace on issues that concern them. Related: Biden’s cryptocurrency framework is a step in the right direction And, it’s not a standalone case. The Securities and Exchange Commission is vying with the CFTC for authority over the world of Web3. Crypto libertarians would dispute whether centralized authorities should have a say at all in an ecosystem that they have only attacked and never aided. The Stabenow-Boozman bill, a proposal in the U.S. Senate, would potentially give the CFTC direct oversight of tokens that qualify as digital commodities. This means that exchanges and online Web3 providers would potentially register with the CFTC, further enmeshing decentralized finance (DeFi) within a centralized web that it was engineered to escape. Monitoring wallets, targeting smart contracts and more The SEC has traditionally sought to regulate cryptocurrency as much as possible. The agency plays a useful role as it is able to pursue instances of outright fraud and Ponzi schemes, which are rampant in Web3. But, there is a stark difference between going after instances of fraud and regulating or governing the industry with regulations that are inapplicable. There are too many question marks related to crypto regulation. One example is related to microtransactions and airdrops. Such transactions take place on many different exchanges over many years, with various price fluctuations. This is impossible to report on from a tax perspective, especially when many platforms are no longer operating. Along with rewards for staking and even derivative tokens liquid staking, it becomes almost impossible to account for. The Biden administration is even targeting Proof-of-Work (POW) blockchains with new “comprehensive guidelines” issued in September. That’s at the same time many administration officials seem to be pushing for a digital USD. Another extremely controversial, draconian crypto regulation that lawmakers have floated includes forcing receivers to verify the personal information of senders when transactions exceed $10,000. They are also seeking to regulate smart contracts as future contracts. And criminal charges are being introduced for those who develop mixers or privacy coins. Though nobody has really said it, what we seem to be witnessing is a war on crypto cloaked in democratic language. The very pillars upon which distributed ledgers have been built are crumbling if these measures are enforced. More conflict to follow? The conflict between traditional regulators and modern finance seems to be reaching a melting point. Regulations are not adapting to meet the needs and strengths of modern DeFi. As such, there is now a standoff between new Web3 protocols and existing legislation. It is almost impossible to deal with the existing legal system as it is not flexible enough to account for DeFi. Ooki DAO is indeed a bad omen for U.S. crypto developers. And it certainly won’t be the last one. A sleuth of bills and procedures are in place. Paradoxically, such actions are likely to simply encourage developers to create programs that are even more resistant to existing laws. The impossibility of complying with existing legislation can leave them with little other choices. Related: Biden‘s anemic crypto framework offered nothing new In one sense, it leaves U.S. crypto developers in the dark regarding what they should develop. From another angle, perhaps the path forward is quite clear. All protocols moving forward may have to be fully decentralized. This was the premise of the very first cryptocurrency, Bitcoin (BTC). Without a central point of failure, there is nobody to target. Developers will have to work on building ecosystems that are completely separate with no ties to the legacy financial system. Blockchains free of identity and Know-Your-Customer (KYC) requirements are the only possible option if developers want to continue operating on American shores. That’s something they are going to have to recognize sooner rather than later. Masha Prusso is the founder of Story VC, an entity that invests in blockchain startups. Freaky Director Slams Day-And-Date Releases: ‘It Destroyed Us’ – CBR – Comic Book Resources https://dentoncountynewsonline.com/freaky-director-slams-day-and-date-releases-it-destroyed-us-cbr-comic-book-resources/ Following the hybrid premiere of Halloween Ends, Freaky director Christopher Landon slams the day-and-date release model for movies. Freaky director Christopher Landon slammed the day-and-date release model following the premiere of Halloween Ends. “Studios: stop gambling with filmmakers and their movies to try and prop up your fledgling streaming services,” Landon wrote in a Twitter threat. “This happened to me on Freaky and it destroyed us. We worked SO HARD to make a fun movie. Blood sweat and tears. Months away from our families. And for what? They love to use the term: ‘two bites of the apple’ but that’s just another way of saying ‘we’re gonna use your movie as a Guinea pig’ for our streaming service.” Landon went on to say that he “begged the studio” not to do a simultaneous release. “Either circle the wagons and protect it for theatrical or just go all in on streaming,” he said. “Don’t split hairs. At least the Halloween folks were made whole. We got hosed.” Both Freaky and Halloween Ends were distributed by Universal Pictures. Released in 2020, Freaky follows the killing spree of a serial killer known as The Butcher (Vince Vaughn) after he accidentally swaps bodies with a high school student Millie Kessler (Kathryn Newton). Freaky premiered in theaters on Nov. 13, 2020, and was released on VOD on Dec. 4, less than a month after its theatrical release. As for Halloween Ends, it was announced in August that Halloween Ends would debut simultaneously in theaters and on Peacock, just like Halloween Kills in 2021. However, Halloween Ends opened to the weakest box office numbers of the entire Halloween sequel trilogy so far, earning $41.2 million across 3,901 theaters. Comparatively, 2018’s Halloween opened to a record-breaking $76.2 million, while Halloween Kills bowed at $50 million. There has been an ongoing “streaming vs. theatrical release” debate since the start of the coronavirus (COVID-19) pandemic and Landon isn’t the only filmmaker to critique hybrid release strategies from studios. Wonder Woman 1984 director Patty Jenkins and Dune director Denis Villeneuve have both spoken out against this strategy, as both of their films were released simultaneously in theaters and on HBO Max. However, there are some on the side of streaming, including Freaky screenwriter Michael Kennedy. “Do I wish FREAKY was released on 3000 screens at a time people could go safely? Yes,” he said in September 2021. “Am I also happy that people got to see it at home during a terrible surge and smile for 102 minutes? Big yes. And it’s finding new life on [HBO Max]. Streaming isn’t the enemy.” Halloween Ends is now available in theaters and on Peacock. Source: Twitter

Dreaming Of Sleeping In A Castle? Here’s Where To Rent One – TheTravel https://dentoncountynewsonline.com/dreaming-of-sleeping-in-a-castle-heres-where-to-rent-one-thetravel/ Our ideas of a great vacation are as different as our ideas about a healthy diet. To some, it’s backpacking while camping in some remote forest jungle like the Amazon. For others, it’s the beach, the sunsets, the stunning views. Yet others can’t just get enough of museums where stories like that of the Titanic, as well as human genius, are re-told—on one side—or exhibited. But whether one loves backpacking, the beach, or museums, a vacation that involves seeing a medieval castle, perhaps even entering one is a surreal experience. Sleeping where powerful monarchs once slept and sitting where they sat can be an experience of a lifetime. In some of these castles, decisions were made that would change the course of history forever. But there were scandals as well. It’s possible to have this kind of vacation. In this article, we reveal some famous castles in Europe that vacationers can rent. Kilkenny Castle: Ireland’s Best-Kept Secret Steeped in fascinating history, the 15th-century Kilkenny’s Tubbrid Castle offers visitors the rare chance to live like a medieval king. In AD 942, and in an attempt to avenge his allies who had been attacked, Muircheartach, a king of Munster, one of the Five-Fifths of Ireland, and later self-declared High King of Ireland marched his troops over to Kilkenny which sat right at the edge of enemy territory. He would rest here and relax before embarking on his military offensive. The Circuit of Ireland—a poem immortalizing this battle—survives. However, the construction of a castle on the site where Muircheartach rested would wait for about 500 years when in the 15th century, Margaret FitzGerald, Countess of Ormond—and a member of the powerful and celebrated FitzGerald dynasty—constructed the castle, even sleeping in it at one time to protect herself from enemy attacks. Defying time and tide, the castle holds over 600 years and more—in its regal walls. Just before its full restoration, it had gone out of use for over 100 years. Today, it combines the old-world look and appeal—with conveniences of modern life like central heating—which is quite rare for a castle, let alone a medieval castle. It’s quietly serene with magical views on all sides. Where Is Tubbrid Castle Located? Tubbrid Castle is 20.3 kilometers, or about a 20-minute drive from Kilkenny City—on the western side via Lacken. While visitors may spend upwards of $1,662 for a minimum 2-night stay at this castle, the cost per head can dip to as low as $208, depending on the number of guests. This is an amazing place to indulge the inner prince. According to some accounts, this castle was built in AD 919. That means visitors to this secluded gem will have over 1,100 years of history to explore, admire—and just gaze at. Llaés Castle is set in the backdrop of lofty mountain peaks and has a striking appearance of a fortress. While many castles are usually quiet and serene, this castle is ridiculously quiet—even if birds occasionally break the almost unnerving silence. The views around are to die for. Stretched out before the eyes are craggy mountain tops or lush pastures where one can spot shepherds tending their flock of sheep in the distance. Other buildings in this castle complex include the tribute tower, the Saint Bartomeu Church, a small cemetery, and the courtyard of arms. While this castle doesn’t offer the premium luxury of a Ritz-Carlton or a Waldorf Astoria—and has some amenities that don’t look modern—a stay here will still be exciting just for the experiences like taking local wines on the balcony while admiring the forested mountains all around. The castle has eight double rooms (five of which have double beds) and has a capacity to accommodate 16 guests or even 21 with auxiliary beds. Visitors can rent the entire castle, rent the 10-person Tower Apartment, or the 6-person School Apartment. While one will need $1,350 to rent the entire castle, the School Apartment goes for as little as $400. Finally, this castle is in Llaés, 15 km from Ripoll. The Positano Watchtower On The Amalfi Coast Blends The Old And The New To fend off attacks by the feared Saracens as well as pirates, Normans would build a guardian tower on a promontory along the stunning Amalfi shoreline in the 13th century. Also called Silja Tower or “Torre Silja,” this castle has been redesigned so that it almost looks like a 21st-century building. It sits literally by the sea, is a short walk from Positano Town and can house up to eight guests. How Much Is Accommodation At Silja Tower? Accommodation for the minimum 2-night stay duration at Silja Tower is $3, 371 for the two days. Finally, this castle is located in the incomparable Positano, one of Italy’s most romantic vacation destinations. That’s it. While we may not be kings, we can still live as kings—literally. And for that, there’s no better place to stay than in a medieval castle.

Business Council Of Canada Says Nexus Closure ‘deeply Troubling’ In Letter To U.S. – CTV News https://dentoncountynewsonline.com/business-council-of-canada-says-nexus-closure-deeply-troubling-in-letter-to-u-s-ctv-news/ OTTAWA – The Business Council of Canada says it is concerned over the continued closure of the Nexus trusted-traveller program, which allows pre-screened travellers expedited processing when entering the United States and Canada. CEO Goldy Hyder says it is “deeply troubling” that the U.S. government has not reopened 13 Nexus enrolment centres, in a letter to David Cohen, the American ambassador to Canada, obtained by The Canadian Press. The two countries are in dispute over a long-standing request by the U.S. Customs and Border Protection agency that its agents be afforded the same legal protections inside Nexus facilities in Canada that they currently have at ports of entry like airports and the Canada-U.S. border. Public Safety Minister Marco Mendicino has cited the principles of Canadian sovereignty in explaining why U.S. customs officers can’t have the same legal protections at Nexus centres that they do at airports and the border. Hyder says in his letter to Cohen that he fears the dispute will hurt businesses whose employees do not already have a Nexus card and he strongly urges the ambassador to recommend reopening the enrolment centres. His comments come on the heels of Canada’s envoy to the U.S. saying the program is being “held hostage” by unilateral American efforts to renegotiate the preclearance agreement between the United States and its northern neighbour. “There’s an attempt to renegotiate the terms of a 20-year-old program unilaterally and the program is being held hostage to that effort,” Kirsten Hillman told a symposium on the Canada-U.S. border hosted last week by the Future Borders Coalition. This report by The Canadian Press was first published Oct. 16, 2022.

Marathon Runner On Living And Running With MS Prevention Magazine
Marathon Runner On Living And Running With MS Prevention Magazine
Marathon Runner On Living And Running With MS – Prevention Magazine https://dentoncountynewsonline.com/marathon-runner-on-living-and-running-with-ms-prevention-magazine/ I started training for my first marathon in 2016. I’m competitive by nature, so I had high expectations of myself and was excited to show off my skills to my family in my hometown of Chicago. But things didn’t go as I’d planned. During the race, I had trouble lifting my foot. I stopped for a while in the medical tent and ended up walking, rather than running, to the finish line. It wasn’t the way I’d hoped to finish but I figured that a visit to a doctor in Houston, where I live, would get me through my injury. By the time I went to the doctor, though, I was noticing other symptoms that caught me off guard. For instance, not only was I having trouble lifting my foot (known as foot drop), I was feeling exhausted daily and often I wasn’t able to feel my extremities. The doctor told me I didn’t have a sports injury and that instead I should see a neurologist. After several frustrating months of appointments and testing—including an MRI and bloodwork— I was diagnosed with primary progressive multiple sclerosis, which is a disease that develops when the immune system attacks the central nervous system and causes nerve damage. The “progressive” part of my diagnosis meant that my symptoms would continue to get worse and would drastically limit my functioning. Oddly enough, getting the diagnosis was a bit of a relief because I finally knew the reason I was experiencing tremors, numbness, and fatigue. Of course, I was also scared, but I was referred to an MS clinic near me and I trusted that the doctors there would help me. Living life with MS To my shock, I was told that based on my neurological loss, I would have only about five more years until I’d be confined to a wheelchair. They told me to travel and do all the things I wanted to do while I was still able. Not wanting to think about the future, my husband Joe and I chose to focus on the present and travel to as many places on our bucket list as possible, as quickly as we could. More From Prevention In April of 2019, we took our first bucket-list trip to Ireland. As luck would have it, there was an MS run/walk happening there and we signed up. In the race packet, I read about “HSCT” (technically called hematopoietic stem cell transplantation) for the first time, which was described as an experimental treatment that could stop MS progression by essentially wiping out your immune system then receiving healthy stem cells to “reset” it. There are trials underway here, but it’s not available in the U.S. as a standard treatment option—and it’s extremely expensive, risky, and new. So I put it on the back burner and continued to focus on making the moments I had count. I continued running, but it was getting harder and harder. Even though I didn’t want to acknowledge it, I had to come to terms with the fact that my racing days were limited. I remember telling my running partner that once it became impossible for me to run, I’d just cheer her on. But I was hard headed and wasn’t going to stop without a fight. What my invisible disability looks like I usually didn’t even appear to have a disability to the outside world. In fact, when I was first diagnosed and got the handicap placard for my car, people actually yelled at me for using a handicap space because they didn’t think I had a true disability. That would upset me, but I’m not one to yell out my personal diagnosis across a parking lot to someone who is unimportant to my life, so I tried to ignore that as much as I could. One thing that helped, however, was that I got a therapy dog, named Hope, who was trained through the National Multiple Sclerosis Society to recognize and assist with tremors and seizures. I put a vest on her that said, “I’m visible. Her disability isn’t.” As Joe and I were about to fly to Paris on our second bucket-list trip—where I was planning to run my last half marathon ever—I came to a realization. There I was in the airport, using a cane (albeit a cute one), wearing an e-brace on my leg (a device that sends electrical shocks to help me lift my foot), and watching Joe lug around a big wheelchair the doctors had suggested we bring just in case I needed it. And it struck me that if I didn’t try to stop the progression of my MS, we were headed for a lifetime with that wheelchair and a future where my husband and daughter would have to wait on me hand and foot. So Joe and I decided we had to at least see if I was a candidate for HSCT. The treatment is not for everyone with MS, but I qualified for it, and we hurried to make arrangements to go the HSCT clinic in Mexico in January 2020. I had a lot of fears and there was a 20 to 30 percent chance that the treatment wouldn’t work, but I felt that this was my shot, and I had to take it. Patrick Pressgrove Where I am today The treatment wasn’t easy, but I really did go into the clinic with my cane, and walked out of the clinic a month later on my own. It was surreal. Afterwards, my physical therapist helped me to strengthen the muscles I’d stopped using and retrain my weakened body, and slowly but surely, I learned to walk and run with an improved gait that helps me control my foot. I wasn’t the same runner I was before my MS diagnosis but I was determined to run marathons again because training puts me in a positive mindset and gives me something to work toward. I soon learned that I qualified for the 2021 Boston Marathon as an adaptive athlete (a person with disabilities who is allowed modifications during sports competitions) and I went for it. Unfortunately, during the race, I had to stop at mile 20 because I suffered an extreme bout of foot drop. I had to move forward carefully so as not to cause further damage, which is a risk when the muscles are strained. I was disappointed that I had to walk the rest of the way to the finish line, but the experience led me to an even more exciting opportunity: I was picked by Degree Deodorant to get another chance at finishing a marathon as one of their “Not Done Yet” runners for the San Francisco Marathon in July 2022. I was thrilled because being a part of the campaign would enable me to spread the word about HSCT and show others that those of us with disabilities can break barriers. I trained hard for this one. And it paid off. When I ran across that finish line, it was momentous. It was definitely an “ugly cry” moment. Now, I’m ready for more. I’m back in training for my next marathon. I still have tremors when my muscles are fatigued. I also have to be very particular in my movements in order to do things that were once simple, and that takes a lot of extra effort and focus. But I push through it because it’s worth it and because I’m not going to let MS define me. I might stumble but I don’t quit, and I hope that inspires other people with disabilities to go for their goals, too. Back to the We Are Not Invisible project

Kelce33 REC, 347 YD, 7 TDS See New Posts Pinned Harrison Butker, good from 62 at the end of the first half, can’t connect on a 51-yarder to cap an 11-play drive by the Chiefs. That was one of Kansas City’s most methodical series of the game, but the kick goes wide left, and it’s still 10-10. Bills will start from their own 33, which means they’ll be well positioned to get their own points on their first drive of the second half. Pinned Mirror image of the last Bills-Chiefs matchup, from the playoffs: K.C. has just over 10 seconds to operate before the break, and Mahomes hits two quick throws to get his squad in field goal range. And then Harrison Butker drills a 62-yard shot to tie it up at 10! Crazy. We go into halftime at 10-10, and this truly has been a dead-even showdown. Pinned Wow. The big-play gambles finally pay off for Buffalo. Josh Allen finds Stefon Diggs downfield to put the Bills in Chiefs territory, and then he lofts a perfect long ball to Gabe Davis. The 34-yard throw was on the money, and Davis just straight-up beat rookie Joshua Williams down the sideline. Not the first time Davis has torched the K.C. secondary. It’s now 10-7 in favor of the visitors. Pinned Back-to-back pressures from Von Miller and Matt Milano contain Mahomes, forcing a Chiefs punt. Suffice to say, it’s been a low-scoring affair so far. Neither side is completing a high percentage of throws, and yet they continue to lean on the downfield game. Isaiah McKenzie, meanwhile, is having an absolute nightmare of a game so far, dropping a wide-open pass from Allen that would’ve brought the Bills out from their own end zone. He’s now had multiple drops and was part of the fumbled exchange with Allen on Buffalo’s first series. Bills fortunate to escape a safety on their latest possession, with Allen just getting out of the end zone on a second-down scramble. Pinned This is a fun watch by nature of Josh Allen and Patrick Mahomes being the QBs, but it’s not really a good watch in terms of smart, efficient football, at least so far. Both sides are content letting their star signal-callers drop 10-15 yards behind the line on every play, so that they can uncork a big shot. When, in reality, Buffalo for example could just keep running Devin Singletary and be in the red zone every series. There’s not a ton of balance here; just sheer gunslinging. That’s why it’s 7-3 and not 20-17. Pinned A Shaq Lawson sack sets up a rare Chiefs three-and-out, but K.C.’s defense steps up on the ensuing drive to pressure Josh Allen on third-and-long, forcing a throw to no one and producing the Bills’ own three-and-out. These two teams can’t stop copying each other in this game. It’s been a mirror image, except the Chiefs have had the one breakaway play. Pinned Bills once again reach the red zone and cannot get the job done. Three straight incomplete passes to end Buffalo’s bid for points there, with the last one right over the middle to Isaiah McKenzie, who fell as he tried reeling in Josh Allen’s pass, which was just slightly tipped. These teams are now a combined 0 for 3 on red-zone tries. Even worse news for Buffalo: right tackle Spencer Brown is headed to the locker room with an injury suffered on that fourth-and-goal misfire. Chiefs still lead it 7-3. Pinned Josh Allen is a dawg for the way he does not shy away from contact — and, in many cases, embraces it. But man, if you’re a Bills fan, you gotta be on edge every time he runs the ball. A few times in this game already he’s powered into a defender near the sticks and taken big shots. Again, it’s part of what makes him so special, but the ultimate key for Buffalo is keeping him upright. Pinned Where is the Bills’ drive to finish a play on defense? It certainly wasn’t evident on the Chiefs’ last few plays. First Patrick Mahomes is driven back in the pocket and forced to throw the ball away, but had Buffalo’s pursuing D-linemen actually kept running at him, he may have been in danger of taking a deep sack. Then, a few snaps later, Mahomes buys time in the pocket and finds JuJu Smith-Schuster, who escapes two half-hearted tackle attempts while in traffic and easily guides into the end zone, basically untouched, for a 42-yard score. Chiefs now on top 7-3. Pinned Isaiah McKenzie is having a rough day so far. Couldn’t corral the unexpected pitch from Josh Allen on the Bills’ opening drive that wiped away a shot at points. Just got smothered by Justin Reid on a third-down throw that left him flat on his back. But we have our first points now, after the Bills hit a field goal to go up 3-0. Tyler Bass, good for 39, and now it’s back to the Chiefs. Pinned One quarter down. 200 combined yards from the two sides, who have had no trouble moving the ball up and down the field. The Bills’ run game is particularly encouraging, feeding off the threat of Josh Allen airing it out. But zero points thanks to each team getting a little too cute/aggressive in the red zone. This one, as predicted, may well play out as if the last possession wins. Pinned Patrick Mahomes starts just as hot as Josh Allen and seemingly finds Marquez Valdes-Scantling for a TD over the middle, threading the needle with a sidearm zip. But then an ineligible-man-downfield penalty negates the score. Mahomes comes right back and puts a back-shoulder TD throw only where Travis Kelce can get it, but Dane Jackson just barely breaks it up at the last second. Von Miller’s impatience on the very next play, drawing a neutral-zone infraction, gives K.C. new life after the near-TDs, but then the Bills’ front stuffs the Chiefs’ short run attempt. On fourth-and-2, Andy Reid goes for it, and Mahomes picks up the first with his own legs. But then we get a second straight red-zone turnover, as Mahomes forces one to the back of the end zone after scrambling. Kaiir Elam comes down with the takeaway, and we’ve still got no score. Pinned Pinned Bills had been rolling on basically a perfect offensive drive to start this game, marching down the field methodically, keeping Kansas City playing off coverage as Josh Allen hit a wide-open Gabe Davis to start 5 for 5 through the air, and Devin Singletary averaged nine yards per carry running through holes. But then Allen pitched one to Isaiah McKenzie with the end zone in sight, and the receiver apparently wasn’t ready for the ball. The fumble is recovered by the Chiefs, and that’s that. One play kills Buffalo’s momentum and wipes a potential TD drive off the board. Pinned Pinned Pinned Pinned Bills open with the ball. Let the games begin. Josh Allen already hearing the deafening crowd noise at Arrowhead. Pinned Pinned Pinned Pinned Pinned Pinned Almost game time at Arrowhead. The Game of the Week, you might say. The last two times these clubs have met, a trip to the AFC Championship or Super Bowl has been on the line. Very intrigued to see how the Chiefs expand their passing offense, assuming Buffalo keys in on Travis Kelce after his four-touchdown showcase on Monday night. Not to say Kelce can be fully eliminated, but if he’s at least more of a priority for the Bills here, that means Patrick Mahomes needs more from JuJu Smith-Schuster, Marquez Valdes-Scantling, Mecole Hardman, etc. Meanwhile, on Buffalo’s side, are we due for yet another massive Gabe Davis day after he broke out against Pittsburgh? You’d think so, if this is a shootout. Pinned Pinned Pinned Pinned Pinned Pinned Bills inactives include WR Jake Kumerow, OL Justin Murray and DT Brandin Bryant, but none more notable than RB Zack Moss, who’s traditionally served as the top complement to Devin Singletary. Now, that role will belong to rookie James Cook, who’s had an uptick in touches the last two weeks, recently breaking loose on a 24-yard touchdown run against the Steelers. Look for Cook to be heavily involved if Buffalo ends up throwing the ball a lot (likely) in Kansas City. See More

Glass Onion: A Knives Out Mystery Closes London Film Festival With Energetic Premiere Deadline
‘Glass Onion: A Knives Out Mystery’ Closes London Film Festival With Energetic Premiere – Deadline https://dentoncountynewsonline.com/glass-onion-a-knives-out-mystery-closes-london-film-festival-with-energetic-premiere-deadline/ The 66th London Film Festival closed Sunday with a lively screening of Rian Johnson’s murder mystery sequel, Glass Onion: A Knives Out Mystery.   Johnson and the film’s A-list cast, including Benoit Blanc a.k.a Daniel Craig, were all in attendance and conducted a brief Q&A onstage following the premiere.  The screening opened, however, with an extended introduction from BFI CEO Ben Roberts, who paid tribute to outgoing BFI Festival director Tricia Tuttle who leaves the BFI after five years in charge of the London Film Festival and over a decade at the body overall.  “I know I speak on behalf of all of us, you are just a fantastic colleague and a really amazing person,” Roberts said of Tuttle, who came onstage and was presented with a bouquet of flowers.  Throughout the screening, the London audience frequently broke out in applause at the film’s surprising cameos and shocking twists, which Johnson was keen to remind the festival audience to protect from people yet to see the film.  “We’re all cool, no spoilers,” he warned before the film started rolling.  During the Q&A, Johnson also had to prompt his actors not to spill any of the film’s twists as their answers were being recorded. Instead, they sidestepped specific plot points and discussed the film’s themes and the expanded scale of the Knives Out universe.  Unlike the first Knives Out film, which was wholly set in and around a New England mansion, Glass Onion is set in Greece and features a vast set of high-tech gadgets, expensive yachts, and a massive real-life piece of glass shaped like an onion.   “All of this started with my love of Agatha Christie and to do what she did with each one of her books, which is something totally different,” Johnson said.  “She wasn’t repeating herself every time. She was not only using new settings and characters but also new narratives, tones, and genres. So I want the audience to know moving forward that that’s what we’re gonna be doing with this.” Later during the Q&A, Daniel Craig, who returns as the famed detective Benoit Blanc in the film, praised Johnson for creating the Knives Out universe and said he would gladly continue to work with the director.  “I’d work with him rest of my life,” Craig said. The Q&A ended with Johnson heaping praise on Janelle Monáe, who plays a lead role in the film and is a crucial part of the film’s shocking twist.  “Without saying why, the degree of difficulty in this role, and the amount of work she put into every scene, I didn’t even appreciate until I put the whole movie together and watched it for the first time,” Johnson said of Monáe. He added: “She’s just extraordinary.” Glass Onion will be released globally on Netflix on December 23, following a theatrical release in select theaters from November 23-29.

Canyon Crest Academy Students Launch First-Ever Crypto Club – Rancho Santa Fe Review https://dentoncountynewsonline.com/canyon-crest-academy-students-launch-first-ever-crypto-club-rancho-santa-fe-review/ A group of young NFT traders and enthusiasts recently created the new Raven DAO Crypto Club at Canyon Crest Academy. Senior Nathan Benhaim founded the club with juniors Geoffrey Duan and Connor Chung and already 150 students have joined. They meet every Wednesday in business/business finance teacher Craig Pollard’s classroom. Free pizza is a plus. “At Raven DAO Crypto Club we’re passionate about NFT investing and educating our friends and classmates on the opportunity to financially benefit from the NFT boom, like we did,” Nathan said. NFTs also known as non-fungible tokens, are unique digital assets that can be traded between users. Last school year, Nathan and a team of advanced business class students created a successful NFT investing company called Oasis DAO, pooling money from investors and making NFT trades. They achieved a 51% gain of the initial investment for its clients. Nathan Benhaim speaks at a recent Crypto Club meeting. (Yamia Benhaim) “As crypto digital currencies have severely dropped values from their all-time high in November 2021, the market provides us one of the best opportunities to enter the space. By creating Raven DAO Club, our goal is to build a community of peers that are excited to learn more about how crypto and the blockchain technology work,” Geoffrey said. “As NFT traders and enthusiasts, we share our experience on how to safely transact NFTs and research future investments for crypto currencies.” CCA club members can learn more about this economic sector and future career pathways. As Connor said, NFTs and the crypto space in general are unique due to the decentralization aspect and they are excited to explore all of the potential applications with the club. “I am so impressed with Nathan, Geoffrey and Connor,” said Pollard. “They have reversed roles with me and now I am the student learning from them.” To learn more, check out ravendao.app/

Russia Is Making Excuses For Failure To Stop US-Made HIMARS In Ukraine – Business Insider https://dentoncountynewsonline.com/russia-is-making-excuses-for-failure-to-stop-us-made-himars-in-ukraine-business-insider/ Russian forces have struggled to counter Ukraine’s use of US-made HIMARS rockets. Some Russians have an explanation: HIMARS has a secret feature that makes it harder to target them. That’s probably bluster meant to distract from Russian military failings, one expert told Insider. Loading Something is loading. Thanks for signing up! Access your favorite topics in a personalized feed while you’re on the go. Why can’t Russia destroy Ukraine’s US-made HIMARS rocket launchers? One Russian defense blog has an explanation: HIMARS has a secret feature that prevents Russian artillery from targeting it. Not quite, Western defense experts say. The more likely reason is Russian incompetence. In September, Russian defense blog Avia wrote about why Russian artillery has been not able to knock out Ukraine’s M142 High Mobility Artillery Rocket Systems, which fire GPS-guided rockets that have savaged vital Russian targets such as ammunition dumps, command posts, and bridges. A HIMARS during an exercise in Latvia in September. GINTS IVUSKANS/AFP via Getty Images In theory, Russia’s huge arsenal of howitzers and multiple-launch rocket systems should be able to destroy HIMARS by using counter-battery radar to track the trajectory of the rockets it fires back to their launch point. Ah, but those clever Americans have a trick, according to Avia’s unsigned blog post: They designed HIMARS so its rockets would change trajectory and fool counter-battery radars. “This can be seen from the missile’s flight path, which, in fact, shifts the coordinates set by counterbattery means of combat by hundreds of meters, making it impossible to deliver accurate strikes,” the post says, pointing to videos of Ukraine firing the rockets. “Experts draw attention to video footage published by the Ukrainian military, which shows that after launch, the rocket changes its flight path almost immediately,” the post says, according to a translation done by Google. “This greatly distinguishes American systems from conventional MLRS [multiple launch rocket systems], where the projectiles fly along a ballistic trajectory. With a high degree of probability, it is this fact that prevents the establishment of the exact location of the coordinates of the launchers,” it adds. A HIMARS is fired on the flight deck of the amphibious ship USS Anchorage in October 2017. US Navy/Mass Comm Specialist 2nd Class Matthew Dickinson Do HIMARS rockets really alter their trajectory after launch, in the same way that NASA rockets pivot their engines to change course during their ascent to orbit? In response to a query from Insider, HIMARS manufacturer Lockheed Martin deferred comment to the US Army. The US Army’s response was simply that “the missile attains the ballistic trajectory it’s assigned to reach the target.” Counter-battery fire — using artillery to knock out other artillery — is a difficult process even under the best of circumstances. But poor Russian counter-battery capabilities are compounding the problem. “The article is probably grasping at straws,” Samuel Cranny-Evans, a land-warfare expert at Britain’s Royal United Services Institute think tank, told Insider. For example, Russia’s Zoopark-1 counter-battery radar can detect rockets at a ranges of 9 to 13 miles, but HIMARS rockets have a range of about 50 miles. “The radar needs to pick the projectile up as it is launched to try and predict its trajectory and plot its likely approach before extrapolating backwards to reach a probable launch point,” Cranny-Evans said. “So if it doesn’t pick the rocket up from its launch point because it’s not within range, it can’t provide a targeting solution.” US soldiers set up an AN/TPQ-53 Q-53 counter-fire target acquisition radar during an exercise in Hawaii in June. US Army/1st Lt. Steph Sweeney In addition, counter-battery radars are set to scan for incoming shells and rockets passing through a specific sector at a specific height. “Unless the radar happens to be looking in the right place at the right time, it will not detect a HIMARS launch,” Cranny-Evans said. “I doubt the Russians have enough counter-battery radars to provide continuous coverage and so would be limited in their ability to provide persistent monitoring even if they could get within range of the rockets.” Counter-battery radars also give an approximate location of the firing battery. While this may be good enough to lay down a general barrage and hope to hit something, it’s not accurate enough for a precise shot. This is especially true for mobile artillery, such as truck-mounted HIMARS and armored self-propelled howitzers, that use “shoot and scoot” tactics to fire a salvo and relocate within minutes. That tactic requires counter-battery fire to be launched within minutes of detecting incoming fire – and Russia’s command structure has been too slow to do this. Ukrainian troops with a captured Russian self-propelled gun in Izium on September 14. Viacheslav Mavrychev/Suspilne Ukraine/JSC “UA:PBC”https://www.businessinsider.com/Global Images Ukraine via Getty Images “The Russians appear to have a very slow targeting process that is often unable to take account of moving targets or a changing situation,” Cranny-Evans said. “If they do not immediately fire on the suspected HIMARS location, or the fire mission gets allocated a low priority for whatever reason, they will not be able to engage.” While this Avia article appears groundless, Russian defense blogs — which often have links to the Russian government and military — can be quite illuminating. The HIMARS article reveals the depth of Russian frustration with Ukraine’s new Western-supplied weapons. When Russian forces invaded in February, their more modern artillery outranged Ukraine’s older Soviet-era arms. Now the shoe is on the other foot. The article also suggests that someone in Moscow is looking for scapegoats. Rather than fixing poor equipment or ineffective procedures, they find it easier to blame defeat on enemy secret weapons. Tech News: 5 Things To Know In Australia Today – Gizmodo Australia https://dentoncountynewsonline.com/tech-news-5-things-to-know-in-australia-today-gizmodo-australia-6/ Hello, hope you had a good weekend. There are a few tech things to catch up on this morning, so we’ll get straight to them. 1. Woolworths subsidiary suffers data breach Online marketplace MyDeal, which is a subsidiary of Woolworths, revealed 2.2 million customer records had leaked from its CRM thanks to a ‘compromised user credential’. Data accessed includes customer names, email addresses, phone numbers and delivery addresses, and in some instances, dates of birth (if provided to purchase alcohol). MyDeal does not store payment, drivers licence or passport details and no customer account passwords or payment details have been compromised. No other Woolworths system was involved in the breach. 2. QLD e-scooter penalties ‘designed to hurt’ Queensland Premier Annastacia Palaszczuk announced Friday afternoon that from November 1, tough penalties will apply for those using an e-scooter in the state. The biggest fine will be $1,078 for using your phone while riding, with drink driving the next highest penalty, at $431. The fines are hefty, but state Transport Minister Mark Bailey said, “The fines are going to hurt, they’re designed to”. Be safe out there, folks. These new penalties come into effect from November 1. pic.twitter.com/HuqV30xnal — Annastacia Palaszczuk (@AnnastaciaMP) October 15, 2022 3. An Apple tablet-speaker-hub smart display Bloomberg’s Mark Gurman is reporting his well-backed theory that Apple is working on a dock for the iPad that would allow the device to function as a smart display and speaker. Kind of like Amazon’s Echo Show or Google’s Nest Hub (or its newly touted Pixel Tablet). The product is expected to launch soon, but unlike the usual product drops, this one will be pushed out with a little less fanfare. 4. SpaceX will keep funding Starlink internet in Ukraine After saying he could not keep paying for Starlink satellite in Ukraine, Elon Musk on Saturday literally said “The hell with it” and decided that his company would continue to fund it. He said the loss is around $US20 million a month, which is no small sum. Good to see he’s doing something decent. He could do all this without all the ballyhoo, but. The hell with it … even though Starlink is still losing money & other companies are getting billions of taxpayer $, we’ll just keep funding Ukraine govt for free — Elon Musk (@elonmusk) October 15, 2022 5. Nvidia invents new word (and pulls graphics card) Nvidia is delaunchifying, un-unleasing, decatapulting — OK fine, we’ll use the word the company used — “unlaunching” its 12 GB version of the upcoming RTX 4080 graphics card. Why? Well according to a blog post, the card is “fantastic,” but “it’s not named right. Having two GPUs with the 4080 designation is confusing.” Not helping Nvidia’s attempts to avoid confusing its customers, the company’s short and un-revealing announcement isn’t completely clear about what’s really going on here. BONUS ITEM: Over the weekend, the hearts of many ached. RIP Robbie Coltrane. There’s no Hogwarts without you, Hagrid. Have a good week, folks.

Why These Villages On The Italian Riviera Are Molto Bene! TheTravel
Why These Villages On The Italian Riviera Are Molto Bene! – TheTravel https://dentoncountynewsonline.com/why-these-villages-on-the-italian-riviera-are-molto-bene-thetravel/ Less celebrated than the glamorous French Riviera but every bit as visually stunning, the Italian Riviera calls to travelers uninterested in bright lights, big city destination, and instead favors unique, romantic villages, rocky cliffs, and sandy coves along a stretch of the Ligurian Sea in Italy’s northwest corner. The Italian Riviera begins at the country’s border with France and continues east beyond Genoa, following the scenic coastline southward toward Tuscany. Its best-known destinations include the postcard-perfect Cinque Terre, made up of five small villages, plus Portofino, Ventimiglia, Alassio, Savona, Sestri Levante, Genoa, and Santa Margherita. It’s a region that traditionally attracts celebrities and other wealthy travelers, and its most famous hotel is the luxury Belmond Hotel Splendido, in Portofino, an elegant property high up on a hill and overlooking the town’s picturesque harbor. It has an illustrious guest list that over the years has included Grace Kelly, Walt Disney, and Winston Churchill. And it’s where, according to reports, Richard Burton proposed to Elizabeth Taylor. Here’s what visitors need to know about these Italian Riviera villages. Portofino Is Considered The Jewel Of The Italian Riviera One of the most popular resort areas along the Italian Riviera, Portofino is famous for the colorful homes and buildings that surround its crescent-shaped harbor, which is typically filled with yachts and fishing boats. The vibrant harbor neighborhood has many restaurants and cafés, and luxury stores such as Louis Vuitton and Gucci. The site where the Hotel Splendido now stands was formerly a Benedictine monastery, in the 15th century. It was soon abandoned due to pirate raids and became a ruin until a new owner restored it as a villa hotel in the 19th century. It quickly became a favorite vacation spot for British aristocrats. Travelers can get a sense of the destination’s character by walking through the village, visiting the Piazza Martiri dell’Olivetta, a pretty square located at the harbor, strolling the Via Roma, the town’s main road with many shops and eateries, and exploring the Piazza Della Libertà, the central square in the old town. History buffs should stop at the 12th-century Church of San Giorgio, and at the 15th-century Brown Castle, a former military fortress now surrounded by a rose garden and offering fantastic sea views. Beyond the ultra-luxury Hotel Splendido, travelers looking for more modestly priced accommodations have several choices in Portofino, including these: Hotel Piccolo Portofino: A historic villa turned waterfront hotel with a private rocky beach and a bar, this hotel offers rates around $182 per night. Albergo Nazionale: Located directly on Portofino harbor, the hotel features great views and nightly rates of about $230. Eight Hotel Portofino: In a sought-after location between the harbor and the surrounding mountains, this upscale property has nightly rates in the $600-$650 range. B&B Tre Mari Portofino: Found in the foothills of Portofino, this bed-and-breakfast establishment can be booked for about $170 per night. Cinque Terre Features Five Destinations In One Cinque Terre’s five villages (sometimes perceived as tourist traps) are Monterosso al Mare, Vernazza, Corniglia, Manarola, and Riomaggiore, and all are located within the Cinque Terre National Park, a UNESCO World Heritage Site. Monterosso al Mare, the largest of the five, is a hilltop region known for its lemon and olive groves, and its beaches. The ruins of the Middle Ages-era Castle Monterosso dominate the old town, while a sculpture of Neptune marks the new town, called Fegina. Vernazza is a small fishing village considered one of the most beautiful towns in all of Italy. It has a medieval castle, Doria Castle, built in the 1500s, and its piazza features restaurants, cafés, and bars. Corniglia is an inland village, dating to Roman times, without ocean access and is an agricultural area surrounded by vineyards. Manarola is another hilltop village with steep roads and alleys leading to the waterfront. Its small harbor features the pastel-colored houses the region is known for, and its modest piazza offers a few seafood restaurants. Riomaggiore, the southernmost village of the Cinque Terre region, offers great ocean views, stone houses and buildings, and the 11th-century church Santuario di Nostra Signora di Montenero. Other sights to see in Riomaggiore include the Castle of Riomaggiore, built in 1260, and the gothic Church of San Giovanni Battista, dating from 1340. A 7.5-mile footpath, called the Blue Path, connects all five villages and is the best way to experience each of the destinations up close. Cinque Terre Offers Some Of The Best Beaches There are beaches and coves at nearly every turn along the Cinque Terre coast. Most of the bigger beaches will have public and private areas, with the private areas often charging a fee but offering services such as water sports rentals and places to eat, plus showers and restrooms. Monterosso has a popular beach in the new part of the village. A beachside promenade is lined with restaurants, pubs, and ice-cream shops, and visitors can rent chairs and umbrellas at the beach. Bonassola Beach, nearby Monterosso, is one of the largest bathing beaches in the region and is great for swimming and snorkeling. Chairs and umbrellas can be rented and there is food service. Manarola Harbor offers deep-water swimming. A ladder straddling the harbor’s rocky cliffside is provided for adventurous swimmers. Riomaggiore offers a secluded and rocky beach near the town center. Its pebble beach isn’t the best for sunbathing, but the beach is considered a nice swimming spot. Water shoes are recommended.

Ghana’s Resolve To Halt Galamsey Unprecedented – Akufo-Addo – Ghana Business News https://dentoncountynewsonline.com/ghanas-resolve-to-halt-galamsey-unprecedented-akufo-addo-ghana-business-news/ President Akufo-Addo The national campaign to halt illegal small-scale mining (galamsey) is unprecedented as most well-meaning Ghanaians have thrown weight behind the initiative, President Nana Addo Dankwa Akufo-Addo said on Sunday. “Everybody is talking about it and that connotes the magnitude of the problem. The government’s agenda is to involve all identifiable groups in dealing with the issue head-on,” he said. President Akufo-Addo, who paid a courtesy call on the Asantehene, Otumfuo Osei Tutu II, at the Manhyia Palace in Kumasi, rallied the citizenry to give their all to make the anti-galamsey crusade successful. The President is on a five-day working visit to the Ashanti Region to inspect and inaugurate development projects. Projects billed for inspection include the Komfo Anokye Teaching Hospital Maternal and Children’s Block, Kejetia/Central Market Phase Two, Obuasi Trauma and Accident Hospital, and Boankra Inland Port projects. The rest are the Anwiankwanta-Obuasi and Santasi-Apire Road, Oforikrom, Nsuta and Trede Agenda 111 projects, as well as a sod-cutting ceremony for work to begin on the Suame Interchange. President Akufo-Addo is also expected to inaugurate the Toase Magistrate’s Court, residential accommodation for the Appeals Court judges, the ECG KATH 33/11 kv Primary sub-Station, and Nsuta Maize Processing Factory, which had been completed under the ‘One District, One Factory programme. On infrastructural projects executed in the region over the last six years, the President cited the construction and rehabilitation of about 400 kilometres of asphalted roads, as well as over 30 hospital projects meant to make life comfortable for the people. The President said the Government had worked assiduously to impact positively on the living conditions of the people and determined to improve upon it. Otumfuo Osei Tutu, welcomed the President to the region and lauded him for regulary visiting to inspect ongoing development projects. That, he said, was necessary to ensure the successful completion of those projects. Source: GNA

Uncollected Solid Waste Posing Health Hazards To Multanites – Pakistan Today https://dentoncountynewsonline.com/uncollected-solid-waste-posing-health-hazards-to-multanites-pakistan-today/ MULTAN: Solid waste management is getting tougher in the fifth largest metropolitan of the country adding to environmental pollution and endangering hundreds of thousands dweller to serious health hazards. With an urban population of just over 2.1 million, the city daily generates around 1300 tone solid, industrial and hospital waste of which the local authorities have the capacity to only collect only 700 tone waste. The only landfill site used by Multan Waste Management Company (MWMC) since 2013 is now filled up to the brim, leaving MWMC staff with no option to identify another place to dump the waste. The concerned authorities have described the shortage of staff and resource constraint behind uncollected waste as the situation is getting tougher with every passing day due to population influx as well as proper mechanism of waste segregation and treatment. “We have the capacity to collect only 700 tone waste daily against a daily bulge of 1300 tone,” said Manager Operations MWMC Anwar-ul-Haq. “The company is covering 68 urban Union Councils housing over two million people with only 2200 staff including 1500 sanitary workers.” “Expecting 100 percent results with such staff strength was unjust,” the Manager Operations said, revealing that no recruitment of workforce or acquisition of new machinery was made since 2013. ‘The Hospital Management Rules’ were introduced in 2005 necessitating yellow-bagged waste to be disposed by burning and then dump it at a landfill site or dispose it of through any other method approved by the concerned Federal or Provincial agencies. In 2012, the Punjab government had established Waste Management Companies in big cities for swift cleanliness arrangements in the major cities but the objective is yet to be fully achieved in case of Multan due to certain factors despite MWMC fully operational since 2014. Keeping in view the urgency, the Manager Operation said that the company Board of Directors (BoD) recently approved recruitment of 500 workers and procurement of over 100 vehicles at a cost of Rs 170 million in first phase. “These measures would help evolve a swift sanitation system to ensure proper waste disposal.” Although, the officials are busy locating an alternate place to use landfill site, yet there are other issues like hazards of dumping waste in open, proper segregation of house, industrial and hospital waste and having different landfill sites for it. Lack of professional human resource to collect and properly dump or treat solid waste is yet another reason contributing to environmental degradation and hazards to human health. Chief Executive Officer (CEO) MWMC Muhammad Farooq Dogar said that international standards require a sanitary worker deployed for every 1000 persons for proper sanitation and waste management. “This is missing in our case. To fill the gap of company’s limitations, special campaigns are launched periodically targeting specific areas to cover maximum, if not 100 per cent.” Moreover, he said, there was no practice of solid waste segregation to manage different kinds of waste properly. “Waste segregation would ensure proper management and disposal in accordance with the nature of waste.” Officials are busy these days, in searching a new landfill site and locations including 17-Kassi, Labar Morr and Head Muhammad Wala are in focus. A plan is also under consideration to develop a park at Habiba Siyal landfill site to avoid environmental pollution. Talking of equipment, Farooq Dogar said the waste collection containers have completed their life span running for nine years instead of three years specific period. He pleaded to raise the workforce number to 3500 besides procurement of more machinery to cover maximum area of the urban population. He also proposed to enhance the number of present 12 transfer stations to 20 where primary waste is gathered for onward transportation to dumping site. “It is our utmost endeavor to collect maximum waste generated daily,” he said. “Our mission is to provide healthy living conditions and better environment to Multanites.” Assistant Director Environment Hameed Akhtar said, environment should be made part of curriculum to raise awareness on the issue right from primary level. He said, since there is no mechanism in vogue in the city to segregate waste, there is always risk of diseases outbreak due to dumping infectious and hazardous industrial and hospital waste in open. Meanwhile, the residents have called upon the civic agencies to ensure proper collection and disposal of waste also urging residents to cooperate with Waste Management Company. “It is our collective responsibility to keep our city clean. The people should cooperate with sanitary workers and throw the waste only into the containers or places designated by the company,” said a citizen Shafiq Bhatti. Another citizen Tanvir Shakir suggested community participation in the whole exercise. “The waste management company should divide city into wards and engage local notables to educate people in their respective area in cleanliness.” He said that citizens should also hand over the waste to sanitary workers instead of throwing it on roads or streets.

Toward An Inclusive Metaverse: Meta Supports Research Project – MIXED Reality News https://dentoncountynewsonline.com/toward-an-inclusive-metaverse-meta-supports-research-project-mixed-reality-news/ Image: RODNAE Productions (Pexels) The metaverse should also work for people with disabilities. Meta supports a research project that aims to advance the inclusive metaverse. The “Social Virtual Reality” project is concerned with increasing accessibility in VR for people with disabilities, senior citizens, or those impaired by illness. In particular, the group looks at social VR apps such as VRChat, Rec Room, or Horizon Worlds. Social VR can improve quality of life “The buzzing world of SVR has the potential to open up a wide array of opportunities for disabled and older people to engage with life and improve its quality,” explains project leader Dr. Vanja Garaj. The Brunel Design School research director sees great potential in social VR apps. In addition to simple entertainment, for example, social VR combats loneliness. Improving employment prospects for people with disabilities through VR training or VR online education is also possible, she says. A framework for an inclusive metaverse The research team aims to develop a “Social Virtual Reality Inclusion Framework” within the next three years. This will include tools and guidance on how to make the metaverse more accessible for elders, people with vision or hearing loss, dexterity or mobility issues, mental illness, cognitive impairments, or learning disabilities. One such approach is already offered by the VR app WalkinVR, which makes current VR hits such as Beat Saber or Job Simulator accessible to people with physical disabilities. The VR fitness app FitXR also focuses on inclusion, offering workouts for people with mobility impairments, for example. Kündigung jederzeit online möglich ab 2,80 € / Monat Meta supports research for more inclusion in the metaverse Other organizations involved in the project include Cambridge University and tech giant Meta. According to Dr. Garaj, Meta played a decisive role in the realization of the project. Meta’s support helped justify the project at the proposal stage, he said. It is unclear what the group’s involvement was in concrete terms. However, it is likely to be mainly advisory and offering technical support, as the project is funded by the Engineering and Physical Sciences Research Council (EPSRC) to the tune of just under $1.1 million. “We see Social VR as a significant step towards the ‘full-blown’ metaverse and Meta is leading the efforts to establish this new global network,” Garaj said. NFT Bargain-Hunting Is In As Crypto Bust Craters Prices – HT Tech https://dentoncountynewsonline.com/nft-bargain-hunting-is-in-as-crypto-bust-craters-prices-ht-tech/ NFT sales plunged 67% sequentially in the third quarter as speculators and celebrities made an exodus. A year ago, Daniel Maegaard, one of the higher-profile collectors of nonfungible tokens, was spending tens of thousands of dollars on CryptoPunks featuring funky-looking people or apes. In the last few months, he’s been buying NFTs that typically sell for less than $2,000. The shift is indicative of the metamorphosis taking place in the market for the once-coveted digital images promoted by the likes of Justin Bieber and Madonna. NFT sales plunged 67% sequentially in the third quarter as speculators and celebrities made an exodus, leaving the sector mostly to the dedicated long-term believers in the various potential use cases, and collectors betting that the plunge in prices will rekindle speculative demand. “There’s more upside potential compared to other NFT projects with much higher market capitalizations,” said the 32-year-old Maegaard, who began investing in NFTs connected to land sales in the blockchain-based game Axie Infinity in early 2019. Cheaper NFT collections and $100 gaming NFTs have been generally selling better than the expensive stuff, though Maegaard said he did sell a CryptoPunk depicting an ape in a hoodie for $4.45 million on Sept. 28. On average, an Ethereum blockchain-based NFT fetched $120 on Oct. 3, compared with $1,631 in early February, according to market data researcher NonFungible. On the Ronin blockchain used by Axie Infinity, average prices were down to $16 as of Oct. 3, from $69 in February, the researcher found. The steep decline in cryptocurrency prices overall this year is credited mostly for the decline. On top of that, there is mounting concern about whether NFTs are likely to be viewed as securities, in light of the US Securities and Exchange Commission’s investigation of Yuga Labs Inc., the creators of the popular Bored Ape Yacht Club tokens. “It is definitely not a super interesting space for short-term trading these days,” said Gauthier Zuppinger, co-founder of NonFungible. “Which may be an explanation why the space is currently at a pretty low pace.” That’s transforming the competitive landscape, especially for NFT trading platforms, as buyers search for bargains and many new collections debut outside of the Ethereum blockchain. OpenSea, the world’s biggest NFT marketplace, had a 38% market share in September, down from nearly 85% in March, according to tracker DappRadar. Rival platforms x2y2 and Magic Eden have boomed — with Magic Eden more than doubling its sales in September. Magic Eden had a 13.4% market share in September, up from also less than 1% in March, according to DappRadar. Last month, x2y2 boosted its share to 31.3%, up from less than 1% in March. One reason for Magic Eden’s success: the average price of an NFT there is $43, according to DappRadar, versus $82 on OpenSea over the last 30 days. Folks are more apt to scoop up cheaper tokens as they play games, instead of looking to invest thousands or even millions into art they hope will double in price some day. “The NFT market is showing signs of evolution, where the NFT traders went from degens” — a slang word for crypto believers — to more rational investors, said Pedro Herrera, head of research at DappRadar. “There is a highly competitive environment across marketplaces.” In the week of Sept. 19, mints of new NFTs reached all-time highs, according to researcher Nansen — largely due to debut of small NFT collections with low prices. But, Nansen cautions, that doesn’t mean that large, expensive collections are out for good. An index of blue-chip NFTs like Bored Ape Yacht Club, Azuki, CryptoPunks and Doodles still often outperforms others, said Martin Lee, a research analyst at Nansen. “So while average transactions might be lower in dollar/ETH value, a large bulk of attention and value accrual still resides within the more expensive, well-known collections,” Lee said. What has changed, perhaps, is the investor mindset. “The mania of paying outrageous prices for unexceptional NFTs is gone for good,” said crypto investor Aaron Brown, who writes for Bloomberg Opinion. “I see potential appreciation for NFTs with substance, although I expect NFTs to show the same kind of ups and downs, fads and forgetfulness, of all collectible markets.”

OPEC+ Members Line Up To Endorse Output Cut After U.S. Coercion Claim – Reuters https://dentoncountynewsonline.com/opec-members-line-up-to-endorse-output-cut-after-u-s-coercion-claim-reuters/ U.S. claimed more than one OPEC country coerced into cut Iraq Kuwait, other OPEC+ members, stand by decision Saudi defence minister says decision was purely economic CAIRO Oct 16 (Reuters) – OPEC+ member states lined up on Sunday to endorse a steep production cut agreed this month after the White House, stepping up a war of words with Riyadh, claimed Saudi Arabia had coerced some other nations into supporting the move. Washington noted on Thursday that the cut would boost Russia’s foreign earnings and suggested it had been engineered for political reasons by Riyadh, which on Sunday emphatically denied it was supporting Moscow in its war with Ukraine. The kingdom’s defense minister, Prince Khalid bin Salman, also said the Oct 5 decision to reduce output by 2 million barrels per day – which was taken despite oil markets being tight – was unanimous and based on economic factors. Register now for FREE unlimited access to Reuters.com His comment was echoed by Iraq, OPEC’s second largest exporter, and several other producer states. “There is complete consensus among OPEC+ countries that the best approach in dealing with the oil market conditions during the current period of uncertainty and lack of clarity is a pre-emptive approach that supports market stability and provides the guidance needed for the future,” Iraq’s state oil marketer SOMO said in a statement. Kuwait Petroleum Corporation Chief Executive Officer Nawaf Saud al-Sabah also welcomed the decision by OPEC+ – which includes other major producers, notably Russia – and said the country was keen to maintain a balanced oil markets, state news agency KUNA reported. Oman and Bahrain also said in separate statements that OPEC had unanimously agreed on the reduction. Algeria’s energy minister called the Oct. 5 decision “historic” and he and OPEC Secretary General Haitham Al Ghais, visiting Algeria, expressed their full confidence in it, Algeria’s Ennahar TV reported. Ghais later told a news conference that the organisation targeted a balance between supply and demand rather than a specific price. Oil inventories in major economies are at lower levels than when OPEC has cut output in the past. But some analysts have said recent volatility in crude markets could be remedied by a cut that would help attract investors to an underperforming market. U.S. National Security Council spokesman John Kirby said on Thursday that “more than one” OPEC member had felt coerced by Saudi Arabia into the vote, adding that the cut would also increase Russia’s revenues and blunt the effectiveness of sanctions imposed over its invasion of Ukraine. Khalid bin Salman said on Sunday he was “astonished” by claims his country was “standing with Russia in its war with Ukraine.” “It is telling that these false accusations did not come from the Ukrainian government,” the king’s younger son wrote on Twitter. Updated: 17 Oct 2022, 12:52 AM IST Shouvik Das Premium Inflation and geopolitical tensions, among others, are being cited as key factors for depressed demand. istock Experts say inventories have risen to very high levels due to subdued consumer demand NEW DELHI : Chipmakers such as Taiwan Semiconductor Manufacturing Company (TSMC) and Hon Hai Precision Industry Co., better known as Foxconn, may be forced to rethink their investments in the coming quarters as they take a cautious stance to respond to global headwinds such as rising inventory coupled with concerns of rising inflation and geopolitical tensions. TSMC reported on Thursday an 80% jump in net profit for the September quarter at $8.8 billion. However, even as the results beat market estimates, chief executive C.C. Wei said that net capital expenditure for the rest of FY23 has been cut by 10% to $36 billion. He said also that there could be a “likely decline” in the global semiconductor market, and TSMC is “not immune” from the downturn despite its dominant market position. Foxconn, too, advised caution in its outlook for the rest of the year while announcing its September sales. Even as it reported record monthly revenue of $25.9 billion last month, a sequential rise of 83%, the company cited “the dynamics of inflation, the pandemic, and the supply chain” as the key factors of concern. Industry analysts say that the reduced capex plan among global supply chain makers is not unexpected since inventories have risen to very high levels due to subdued consumer demand. Navkendar Singh, associate vice-president of client devices at market researcher International Data Corporation (IDC) India, said the slowdown projection is in line with tapering demand globally. “Even enterprises are spending cautiously, and orders are getting delayed or cancelled, as companies put a freeze on hiring and rethink growth plans,” he said. The demand slowdown has been projected for the immediate quarters in the global information technology (IT) services sectors, too. While Indian software services majors Tata Consultancy Services, Infosys, HCL Technologies and Wipro posted revenue growth, analysts noted a drop in new and active deals among most IT firms indicating a cautious stance among enterprises. Stakeholders in the Indian semiconductor industry are, however, confident that there would not be a direct impact on India’s ability to attract investments in its fledgling semiconductor industry. Vivek Tyagi, chairman of the India Electronics and Semiconductor Association (IESA) said there could be some slowdown in investments by global chipmakers in the coming quarters. “However, it is important to note that semiconductor investments in the country would not come off the back of cutting-edge chips, which is where the slowdown would be. For instance, the automotive sector in India has a demand for chips that are between 28nm (nanometer) and 90nm die size— and this sector is likely to continue with a simmering demand going forward,” he said. Tyagi also said that the Indian semiconductor industry is presently at the stage of attracting investments for chip making factories, which are largely long-term efforts. “As we see it, the demand slowdown and caution in expansion of investments would remain for another 18 to 24 months, following which the global supply chain would stabilize. India could attract assembly, testing, marking and packaging (ATMP) and outsourced semiconductor assembly and test (OSAT) facilities,” he said. The Indian government’s production linked incentive (PLI) plan for semiconductors offers a 50% cash benefit for firms that invest in semiconductor facilities, without a minimum cap on investments. Tyagi said that this, coupled with state-wise land and employment benefits, could see India continuing to receive investments for the semiconductor sector even if it is at a slightly reduced pace.Rajeev Khushu, advisor and board member of IESA, said Bengaluru already has over 80 companies associated with semiconductor manufacturing. Cartoon Network Could Live On Through Gaming GameRant
Cartoon Network Could Live On Through Gaming GameRant
Cartoon Network Could Live On Through Gaming – GameRant https://dentoncountynewsonline.com/cartoon-network-could-live-on-through-gaming-gamerant/ Cartoon Network has brought some of the most popular and memorable animated series in the last two decades. Though it is a pop-cultural icon, recent corporate events have cast much uncertainty on the network’s future. Although it is unclear whether the beloved cartoon characters will find a future in animation, they may be able to continue to live on through gaming. This doesn’t require expensive production, as the studio has previously demonstrated that its characters are compelling enough on their own to make even the simplest games engaging. Going the gaming route would be efficient, as Cartoon Network already has a foundation to start working off. WarnerBros. Discovery hasn’t officially shuttered Cartoon Network, and the animation network has sent out a Tweet attempting to calm concerned fans. Nevertheless, many fans have analyzed WarnerBros. Discovery’s recent actions and come up with grim predictions. Cartoon Network’s Grim Future Cartoon Network formed an essential part of many people’s childhoods thanks to its roster of memorable cartoons. Though the network has gone through different eras with varying levels of quality, it has managed to maintain a connection with children through franchises like Adventure Time, The Powerpuff Girls, The Amazing World of Gumball, and Dexter’s Laboratory, to name a few. Unfortunately, due to new corporate deals, Cartoon Network’s future hangs in the balance. The merger between WarnerMedia and Discovery, Inc. which was completed in 2022, has had cascading effects on the entertainment world. It appears the new entertainment conglomerate places less value on animation and cartoons, as following the merger, there were company-wide reorganizations that led to the shutdown of the Warner Bros. Global Kids, Young Adults, and Classics studio. It was also reported that WarnerBros. Discovery laid off around 26% of its employees, mainly from the scripted, unscripted, and animation divisions. This has led to many worrying that Cartoon Network’s future may be in jeopardy. Although the network has denied these allegations to calm fans, the future does not look bright. Cartoon Network and Gaming Cartoon Network has always had one foot in gaming, and this could become the key to its longevity. During the early days of the internet, the network entertained its viewers by inviting them to play various Cartoon Network flash games online. None of them were particularly complex, but they featured popular characters and contained addictive gameplay. Many of them have since been removed, though there are plenty of lessons that can be learned from them. Although Cartoon Network does offer mobile games through Cartoon Network Games, its efforts on this front could use more marketing and support. This could be especially effective in letting older fans know that they can experience the fun of older Cartoon Network flash games on their phones today. It would also be worthwhile to feature more games centered on some of the network’s older franchises. Nostalgia has always been a big draw for consumers, and many fans would love to see beloved characters like Johnny Bravo and Ed, Edd, and Eddy live on through gaming. Cartoon Network’s current roster of cartoons features popular hits like Craig of the Creek and Teens Titans Go!, and they could flourish in the gaming world just as well as they do in television. While it may not be time to declare the network’s funeral, it is likely that its future will feature significant change following the WarnerMedia and Discovery Inc. merger. If the network plays its cards right, it does not need to spell the death of its most popular characters, as the gaming community would be more than happy to accommodate them. The Cruise Industry Wants You Back, But Is The Public Ready For A Post-COVID Reunion? – Yahoo Life https://dentoncountynewsonline.com/the-cruise-industry-wants-you-back-but-is-the-public-ready-for-a-post-covid-reunion-yahoo-life/ Of all the industries affected by the COVID-19 pandemic, travel was definitely hit the hardest. And more specifically, the cruise industry, for obvious reasons. In 2020 and 2021, CBS News reported that Royal Caribbean and Princess Cruises, two of the largest cruise lines on earth, combined lost a total of $63 billion in revenue and thousands of jobs. A study published by the National Library of Medicine also concluded that negative perceptions of the cruising industry spread like wildfire on social media during the pandemic, after the Diamond Princess cruise ship became the largest outbreak outside China early in the pandemic. Those sentiments have remained, even after pandemic restrictions on most cruise lines have eased, and the cruise industry, anecdotally, appears to be attempting a resurgence. Will folks start flocking to their nearest port, eagerly awaiting a victory over the coronavirus pathogen with a voyage out at sea? The cruise industry is betting the answer is yes. Lately, the cruise industry has been spending millions on a multi-medium marketing campaign that includes traditional TV and internet ads as well as placements in reality TV shows. Below, we take a deep dive into the cruise industry’s reputation rehabilitation. The Cruise Industry Wants You Back I’ve never understood the appeal of cruises, as being stuck on a giant barge out at sea, with no land in sight, with the public sounds unpleasant at best and downright scary at worst. Add in a deadly airborne virus that’s easily transmissible between anybody within six feet of one another and you’ve got yourself a prime cocktail for sinking cruise ticket sales. However, recently cruises have been attempting a comeback in a big way, and collaborating with some large brands to do it. The Bachelorette’s latest season featured a cruise as its picturesque backdrop with Virgin Voyages, and the show also gave away a cruise to everyone in their live studio audience for the Men Tell All pre-finale episode. God knows how much this partnership cost the cruise line. The Real Love Boat on CBS is a dating reality show that takes place on a cruise, paying homage to the classic scripted TV show from the 70’s. Disney announced their newest addition to the Disney Cruise Line at their D23 conference earlier this summer, The Disney Treasure, and who can deny the fascinating world of swingers on #CruiseTok? Don’t ask me why pineapples are THE symbol used by those open to getting to know others, intimately, on adventures out at sea, I’m not sure. But if it isn’t entertaining to binge-watch this world from afar. All of this begs the question: is cruising cool now? Or is the industry just trying to make you THINK it’s cool, and really it’s the same cesspool of venereal diseases and bad buffets it always was? Cruises are the ex boyfriend who you got the ick from a few years back, whose back and desperate for your attention. What does he have to offer you this time around? We decided to ask an expert who has worked in the industry for decades. virgin cruise ship, are cruises back An Optimistic Outlook on Cruises From an Industry Expert All of this is fine and dandy for some people, but I’m still not convinced a cruise is actually a relaxing vacation with or without COVID-19, and I know I’m not the only one who feels this way. Is the industry optimistic it’ll bounce back as airlines have? We picked the brain of Bob Levinstein, the CEO of CruiseCompete, a cruise booking site that aggregates specials from some of the top cruise lines in the world, to gauge a temperature on the cruise industry as a whole. He explained that CruiseCompete, along with the entire industry, experienced a “major downturn” at the start, and highs and lows throughout the pandemic. “As COVID waves came and went, we were on a roller coaster of hopes and crashes, but we’re finally at the light at the end of the tunnel,” said Levinstein. “Cruise bookings are coming back rapidly, but ships are not yet sailing as full as in a normal year. As a result, there are lots of great deals out there for cruisers right now. Think $50 per person per day for your cabin, all of your meals, and lots of free onboard activities,” said Levinstein. Offering a ton of perks and discounted prices sounds like an industry hungry for customers to come back, we’ll see if it proves to be a lucrative strategy. What’s New in Cruising? I asked Levinstein about all of the cruise-related media spots recently, and whether he sees a renaissance happening within the industry. “Absolutely. Lots of new, innovative ships are starting to sail, with new onboard experiences like roller coasters at sea and new dining and lounge venues,” said Levinstein. “Virgin Voyages offers hip, adults-only sailing. More adventure cruises are offering once-in-a-lifetime experiences in places like Antarctica, the Galapagos, and the Sea of Cortez during whale calving season are capturing travelers’ imaginations.” Okay, even I’ll admit that a cruise to Antarctica sounds amazing, but it’s also one of the only ways to get there if I’m not mistaken. What Should Travelers Know About Cruises and COVID?  I also asked Levinstein about the latest in COVID-19 protections on cruise lines and his messaging was clear: he doesn’t view it as a concern. “Just about all of the COVID-era restrictions are gone. Masks went away months ago. Vaccinated passengers generally have no restrictions at all and unvaccinated passengers can again sail with just a quick test prior to boarding,” said Levinstein. “Even this testing requirement for the unvaccinated has already been dropped by some lines and is likely to go away completely in the next few months.” I also asked him what he would say to folks who are concerned about getting sick on a cruise: “I would say: Think of all of the people you know personally who have had COVID. Ask yourself: how many of those people caught it on a cruise? Zero? Cruises were never the problem; COVID was.” Well there you have it. Cruises are sailing the seven seas once again and it’s up to you whether you’ll indulge in their offerings. Personally, I’ll abstain, although the bumpy return of air travel hasn’t felt like a no-brainer alternative to seasickness during the past few months either. It’s a new world for all of us, but whether or not you wish to travel it by boat with a roller coast, movie theater and Mickey Mouse packed inside is up to you. Scientists Reappraise The Role Of ‘Zombie’ Cells That Anti-Aging Medicine Has Sought To Eliminate – Neuroscience News https://dentoncountynewsonline.com/scientists-reappraise-the-role-of-zombie-cells-that-anti-aging-medicine-has-sought-to-eliminate-neuroscience-news/ Summary: Some senescent (zombie) cells are embedded in young, healthy tissue and promote normal repair from damage, researchers say. Source: UCSF Not all senescent cells are harmful “zombies” that should be wiped out to prevent age-related disease, according to new research from UC San Francisco, which found that some of them are embedded in young, healthy tissues and promote normal repair from damage. Scientists have now seen these cells in action in lung tissue, as well as other organs that serve as barriers in the body, such as the small intestine, colon and skin. When they used drugs called senolytics to kill these cells, injuries to lung tissues healed more slowly. “Senescent cells can occupy niches with privileged positions as ‘sentinels’ that monitor tissue for injury and respond by stimulating nearby stem cells to grow and initiate repair,” said Tien Peng, MD, associate professor of pulmonary, critical care, allergy and sleep medicine, and senior author of the study, which appears in Science on October 13, 2022. Aging cells can both damage and heal Peng said it was understandable that scientists at first viewed senescent cells as purely detrimental. As people age, senescent cells accumulate that have characteristics of old, worn-out cells, including the inability to make new cells. Instead of dying like normal aged cells, they to live on, spewing a cocktail of inflammatory compounds that form the senescence associated secretory phenotype (SASP). These factors are linked to Alzheimer’s disease, arthritis, and other age-related maladies including cancer. The catchy name “zombie cells” was coined for them. Using senolytics that target and kill “zombie cells,” researchers made the exciting discovery that clearing senescent cells from animals thwarted or diminished age-related disease and extended the lifespan of the animals. Thereafter, a boom of activity ensued in research labs and pharmaceutical companies focused on discovering and refining more powerful versions of these drugs. But killing off senescent cells has dangers, Peng said. For one thing, this current study showed that senescent cells also possess the ability to promote normal healing through activation of stem cell repair. “Our study suggests that senolytics could adversely affect normal repair, but they also have the potential to target diseases where senescent cells drive pathologic stem cell behavior,” said Peng. Lighting up senescent cells One major challenge to studying senescent cells is that biomarkers of senescence (such as the gene p16) are often quite sparse, making it difficult to detect the cells. In early experiments, researchers extracted cells called fibroblasts into culture dishes, allowing them to grow and produce enough cells to experiment with, and then stressed the cells with chemicals that induced them to become senescent. But in living organisms, cells interact with tissues around them, strongly affecting the cells’ gene activity. This means that the characteristics of cells growing isolated in a glass dish could be quite different from that of cells in their natural environment. To create a more powerful tool for their studies, postdoctoral scholar Nabora Reyes de Barboza, Ph.D. and colleagues improved on a common technique of fusing a relevant gene—in this case, the p16 gene, which is overly active in senescent cells—with green fluorescent protein (GFP) as a marker that can reveal the location of the cells under ultraviolet light. By enhancing the quantity and stability of green fluorescent protein in these senescent cells, Reyes greatly amplified the fluorescent signal, finally enabling the researchers to see senescent cells in their natural habitat of living tissues. “Zombies” stimulate stem cells shortly after birth Using this highly sensitive tool, the researchers found that senescent cells exist in young and healthy tissues to a greater extent than previously thought, and actually begin appearing shortly after birth. The scientists also identified specific growth factors that senescent cells secrete to stimulate stem cells to grow and repair tissues. Relevant to aging and tissue injury is the discovery that cells of the immune system such as macrophages and monocytes can activate senescent cells, suggesting that inflammation seen in aged or damaged tissue is a critical modifier of senescent cell activity and regeneration. In their studies of lung tissue, Peng’s team observed green glowing senescent cells lying next to stem cells on the basement membrane that serves as a barrier preventing foreign cells and harmful chemicals from entering the body and also allows oxygen to diffuse from air in the lungs into underlying tissues. Damage can occur at this dynamic interface. Using this highly sensitive tool, the researchers found that senescent cells exist in young and healthy tissues to a greater extent than previously thought, and actually begin appearing shortly after birth. Image is in the public domain The team saw senescent cells in similar positions in other barrier organs such as small intestine, colon, and skin, and their experiments confirmed that if senescent cells were killed with senolytics, lung stem cells were not able to properly repair the barrier surface. Leanne Jones, Ph.D., director of the UCSF Bakar Aging Research Institute and Stuart Lindsay Endowed Professor in Experimental Pathology, said Peng’s study is truly significant for the field of aging research, where the goal is to help individuals live longer and more healthy lives. See also “The studies suggest that senolytics research should focus on recognizing and precisely targeting harmful senescent cells, perhaps at the earliest signs of disease, while leaving helpful ones intact,” she said. “These findings emphasize the need to develop better drugs and small molecules that will target specific subsets of senescent cells that are implicated in disease rather than in regeneration.” Additional authors include Nabora Reyes, Maria Krasilnikov, Nancy C. Allen, Jinyoung Lee, Ben Hyams, Minqi Zhou, Supriya Ravishankar, Monica Cassandras, Chaoqun Wang, Imran Khan, Michael Matthay, and Dean Shappard from the Department of Medicine, Pulmonary and Critical Care Division, Peri Matatia and Ari Molofsky from the Department of Laboratory Medicine, Makato Nakanishi of University of Tokyo, and Judith Campisi of the Buck Institute. Science Abstract Sentinel p16INK4a+ cells in the basement membrane form a reparative niche in the lung We engineered an ultrasensitive reporter of p16INK4a, a biomarker of cellular senescence. Our reporter detected p16INK4a-expressing fibroblasts with certain senescent characteristics that appeared shortly after birth in the basement membrane adjacent to epithelial stem cells in the lung. Furthermore, these p16INK4a+ fibroblasts had enhanced capacity to sense tissue inflammation and respond through their increased secretory capacity to promote epithelial regeneration. In addition, p16INK4a expression was required in fibroblasts to enhance epithelial regeneration. This study highlights a role for p16INK4a+ fibroblasts as tissue-resident sentinels in the stem cell niche that monitor barrier integrity and rapidly respond to inflammation to promote tissue regeneration.
