Unfortunately, you might have to settle for one of Apple’s 2022 models, after an auction house in California just let a 2007 iPhone go for a cool $US32,783 – around $53,000 in Aussie terms. LGC Auctions was flogging an original 2007 iPhone that is still factory sealed. The box looks more like a Cards Against Humanity expansion pack than it does an iPhone box. Bidding for the iPhone started at $US2,500 (~$4,000) and at about 10:15 AEDT on Monday, some sucker (yes, I’m jealous) had committed $US32,783 to the cause. When it was first launched in 2007, the iPhone retailed for around $US600. The auction lot description describes the phone as being “one of the most important and ubiquitous inventions of our lifetime” and while we definitely agree, a home deposit (almost) seems like ridiculous money to drop on a gadget that’s all-but a cool paperweight. The 2007 iPhone auctioned off boasts a whopping 8GB of storage, a touchscreen, a 2MP camera (lol) and, get this, a web browser. Honestly this is a fantastic find and it’s great looking back at how much things have changed. For comparison, the iPhone 14 Pro Max has a 48MP camera and will come with 1TB of storage if you want to hand over $2,769. This factory sealed, first-release iPhone is in exceptional condition. It’s particularly apparent in the United States, where the Commodity Futures Trading Commission (CFTC) fueled concerns in September with an announcement that it was imposing a $250,000 fine on a decentralized autonomous organization (DAO), Ooki DAO, and its investors. The fine was particularly ominous, considering DAOs are intended to be “regulation proof.” The CFTC said in its statement on the issue that Ooki DAO’s bZeroX protocol offered illegal off-exchange trading of digital assets. The agency took issue with the fact that the founders, Tom Bean and Kyle Kistner, tried to use the existing bZeroX protocol within the DAO to put it beyond the reach of regulators. “By transferring control to a DAO, bZeroX’s founders touted to bZeroX community members the operations would be enforcement-proof,” the CFTC said. “The bZx Founders were wrong, however. DAOs are not immune from enforcement and may not violate the law with impunity.” The fine is not all that surprising. The CFTC and other regulators are not going to abide by a veil of decentralization. But, there is something within the ruling that is extremely worrying to Web3 lawyers and developers. The agency’s complaint indicated that the voters within a given DAO could be distinctly liable. In other words, no longer will only founders be targeted, as users who take part could also be liable. This is sure to have a chilling effect on turning people away from DAOs and Web3 in general. After all, the whole point is to avoid this kind of targeting and to create new ecosystems where all parties can vote in peace on issues that concern them. The Securities and Exchange Commission is vying with the CFTC for authority over the world of Web3. Crypto libertarians would dispute whether centralized authorities should have a say at all in an ecosystem that they have only attacked and never aided. The Stabenow-Boozman bill, a proposal in the U.S. Senate, would potentially give the CFTC direct oversight of tokens that qualify as digital commodities. This means that exchanges and online Web3 providers would potentially register with the CFTC, further enmeshing decentralized finance (DeFi) within a centralized web that it was engineered to escape. Monitoring wallets, targeting smart contracts and more The SEC has traditionally sought to regulate cryptocurrency as much as possible. The agency plays a useful role as it is able to pursue instances of outright fraud and Ponzi schemes, which are rampant in Web3. But, there is a stark difference between going after instances of fraud and regulating or governing the industry with regulations that are inapplicable. There are too many question marks related to crypto regulation. One example is related to microtransactions and airdrops. Such transactions take place on many different exchanges over many years, with various price fluctuations. This is impossible to report on from a tax perspective, especially when many platforms are no longer operating. Along with rewards for staking and even derivative tokens liquid staking, it becomes almost impossible to account for. The Biden administration is even targeting Proof-of-Work (POW) blockchains with new “comprehensive guidelines” issued in September. That’s at the same time many administration officials seem to be pushing for a digital USD. Another extremely controversial, draconian crypto regulation that lawmakers have floated includes forcing receivers to verify the personal information of senders when transactions exceed $10,000. They are also seeking to regulate smart contracts as future contracts. And criminal charges are being introduced for those who develop mixers or privacy coins. Though nobody has really said it, what we seem to be witnessing is a war on crypto cloaked in democratic language. The very pillars upon which distributed ledgers have been built are crumbling if these measures are enforced. More conflict to follow? The conflict between traditional regulators and modern finance seems to be reaching a melting point. Regulations are not adapting to meet the needs and strengths of modern DeFi. As such, there is now a standoff between new Web3 protocols and existing legislation. It is almost impossible to deal with the existing legal system as it is not flexible enough to account for DeFi. Ooki DAO is indeed a bad omen for U.S. crypto developers. And it certainly won’t be the last one. A sleuth of bills and procedures are in place. Paradoxically, such actions are likely to simply encourage developers to create programs that are even more resistant to existing laws. The impossibility of complying with existing legislation can leave them with little other choices. From another angle, perhaps the path forward is quite clear. All protocols moving forward may have to be fully decentralized. This was the premise of the very first cryptocurrency, Bitcoin (BTC). Without a central point of failure, there is nobody to target. Developers will have to work on building ecosystems that are completely separate with no ties to the legacy financial system. Blockchains free of identity and Know-Your-Customer (KYC) requirements are the only possible option if developers want to continue operating on American shores. Kilkenny Castle: Ireland’s Best-Kept Secret Steeped in fascinating history, the 15th-century Kilkenny’s Tubbrid Castle offers visitors the rare chance to live like a medieval king. In AD 942, and in an attempt to avenge his allies who had been attacked, Muircheartach, a king of Munster, one of the Five-Fifths of Ireland, and later self-declared High King of Ireland marched his troops over to Kilkenny which sat right at the edge of enemy territory. The two countries are in dispute over a long-standing request by the U.S. Customs and Border Protection agency that its agents be afforded the same legal protections inside Nexus facilities in Canada that they currently have at ports of entry like airports and the Canada-U.S. border. Public Safety Minister Marco Mendicino has cited the principles of Canadian sovereignty in explaining why U.S. customs officers can’t have the same legal protections at Nexus centres that they do at airports and the border. Hyder says in his letter to Cohen that he fears the dispute will hurt businesses whose employees do not already have a Nexus card and he strongly urges the ambassador to recommend reopening the enrolment centres. I’m competitive by nature, so I had high expectations of myself and was excited to show off my skills to my family in my hometown of Chicago. But things didn’t go as I’d planned. During the race, I had trouble lifting my foot. I stopped for a while in the medical tent and ended up walking, rather than running, to the finish line. It wasn’t the way I’d hoped to finish but I figured that a visit to a doctor in Houston, where I live, would get me through my injury. By the time I went to the doctor, though, I was noticing other symptoms that caught me off guard. For instance, not only was I having trouble lifting my foot (known as foot drop), I was feeling exhausted daily and often I wasn’t able to feel my extremities. The doctor told me I didn’t have a sports injury and that instead I should see a neurologist. After several frustrating months of appointments and testing—including an MRI and bloodwork— I was diagnosed with primary progressive multiple sclerosis, which is a disease that develops when the immune system attacks the central nervous system and causes nerve damage. The “progressive” part of my diagnosis meant that my symptoms would continue to get worse and would drastically limit my functioning. Oddly enough, getting the diagnosis was a bit of a relief because I finally knew the reason I was experiencing tremors, numbness, and fatigue. Of course, I was also scared, but I was referred to an MS clinic near me and I trusted that the doctors there would help me. Living life with MS To my shock, I was told that based on my neurological loss, I would have only about five more years until I’d be confined to a wheelchair. They told me to travel and do all the things I wanted to do while I was still able. Not wanting to think about the future, my husband Joe and I chose to focus on the present and travel to as many places on our bucket list as possible, as quickly as we could. In the race packet, I read about “HSCT” (technically called hematopoietic stem cell transplantation) for the first time, which was described as an experimental treatment that could stop MS progression by essentially wiping out your immune system then receiving healthy stem cells to “reset” it. There are trials underway here, but it’s not available in the U.S. as a standard treatment option—and it’s extremely expensive, risky, and new. So I put it on the back burner and continued to focus on making the moments I had count. I continued running, but it was getting harder and harder. Even though I didn’t want to acknowledge it, I had to come to terms with the fact that my racing days were limited. I remember telling my running partner that once it became impossible for me to run, I’d just cheer her on. But I was hard headed and wasn’t going to stop without a fight. What my invisible disability looks like I usually didn’t even appear to have a disability to the outside world. In fact, when I was first diagnosed and got the handicap placard for my car, people actually yelled at me for using a handicap space because they didn’t think I had a true disability. That would upset me, but I’m not one to yell out my personal diagnosis across a parking lot to someone who is unimportant to my life, so I tried to ignore that as much as I could. One thing that helped, however, was that I got a therapy dog, named Hope, who was trained through the National Multiple Sclerosis Society to recognize and assist with tremors and seizures. I put a vest on her that said, “I’m visible. Her disability isn’t.” As Joe and I were about to fly to Paris on our second bucket-list trip—where I was planning to run my last half marathon ever—I came to a realization. There I was in the airport, using a cane (albeit a cute one), wearing an e-brace on my leg (a device that sends electrical shocks to help me lift my foot), and watching Joe lug around a big wheelchair the doctors had suggested we bring just in case I needed it. And it struck me that if I didn’t try to stop the progression of my MS, we were headed for a lifetime with that wheelchair and a future where my husband and daughter would have to wait on me hand and foot. So Joe and I decided we had to at least see if I was a candidate for HSCT. The treatment is not for everyone with MS, but I qualified for it, and we hurried to make arrangements to go the HSCT clinic in Mexico in January 2020. I had a lot of fears and there was a 20 to 30 percent chance that the treatment wouldn’t work, but I felt that this was my shot, and I had to take it. Patrick Pressgrove Where I am today The treatment wasn’t easy, but I really did go into the clinic with my cane, and walked out of the clinic a month later on my own. It was surreal. Afterwards, my physical therapist helped me to strengthen the muscles I’d stopped using and retrain my weakened body, and slowly but surely, I learned to walk and run with an improved gait that helps me control my foot. I wasn’t the same runner I was before my MS diagnosis but I was determined to run marathons again because training puts me in a positive mindset and gives me something to work toward. I soon learned that I qualified for the 2021 Boston Marathon as an adaptive athlete (a person with disabilities who is allowed modifications during sports competitions) and I went for it. Unfortunately, during the race, I had to stop at mile 20 because I suffered an extreme bout of foot drop. I had to move forward carefully so as not to cause further damage, which is a risk when the muscles are strained. I was disappointed that I had to walk the rest of the way to the finish line, but the experience led me to an even more exciting opportunity: I was picked by Degree Deodorant to get another chance at finishing a marathon as one of their “Not Done Yet” runners for the San Francisco Marathon in July 2022. I was thrilled because being a part of the campaign would enable me to spread the word about HSCT and show others that those of us with disabilities can break barriers. I trained hard for this one. And it paid off. When I ran across that finish line, it was momentous. It was definitely an “ugly cry” moment. Now, I’m ready for more. I’m back in training for my next marathon. I still have tremors when my muscles are fatigued. I also have to be very particular in my movements in order to do things that were once simple, and that takes a lot of extra effort and focus. But I push through it because it’s worth it and because I’m not going to let MS define me. Instead, they sidestepped specific plot points and discussed the film’s themes and the expanded scale of the Knives Out universe.  Unlike the first Knives Out film, which was wholly set in and around a New England mansion, Glass Onion is set in Greece and features a vast set of high-tech gadgets, expensive yachts, and a massive real-life piece of glass shaped like an onion.   “All of this started with my love of Agatha Christie and to do what she did with each one of her books, which is something totally different,” Johnson said.  “She wasn’t repeating herself every time. She was not only using new settings and characters but also new narratives, tones, and genres. One Russian defense blog has an explanation: HIMARS has a secret feature that prevents Russian artillery from targeting it. Not quite, Western defense experts say. The more likely reason is Russian incompetence. In September, Russian defense blog Avia wrote about why Russian artillery has been not able to knock out Ukraine’s M142 High Mobility Artillery Rocket Systems, which fire GPS-guided rockets that have savaged vital Russian targets such as ammunition dumps, command posts, and bridges. Ah, but those clever Americans have a trick, according to Avia’s unsigned blog post: They designed HIMARS so its rockets would change trajectory and fool counter-battery radars. “This can be seen from the missile’s flight path, which, in fact, shifts the coordinates set by counterbattery means of combat by hundreds of meters, making it impossible to deliver accurate strikes,” the post says, pointing to videos of Ukraine firing the rockets. “Experts draw attention to video footage published by the Ukrainian military, which shows that after launch, the rocket changes its flight path almost immediately,” the post says, according to a translation done by Google. “This greatly distinguishes American systems from conventional MLRS [multiple launch rocket systems], where the projectiles fly along a ballistic trajectory. With a high degree of probability, it is this fact that prevents the establishment of the exact location of the coordinates of the launchers,” it adds. In response to a query from Insider, HIMARS manufacturer Lockheed Martin deferred comment to the US Army. The US Army’s response was simply that “the missile attains the ballistic trajectory it’s assigned to reach the target.” Counter-battery fire — using artillery to knock out other artillery — is a difficult process even under the best of circumstances. But poor Russian counter-battery capabilities are compounding the problem. “The article is probably grasping at straws,” Samuel Cranny-Evans, a land-warfare expert at Britain’s Royal United Services Institute think tank, told Insider. For example, Russia’s Zoopark-1 counter-battery radar can detect rockets at a ranges of 9 to 13 miles, but HIMARS rockets have a range of about 50 miles. “The radar needs to pick the projectile up as it is launched to try and predict its trajectory and plot its likely approach before extrapolating backwards to reach a probable launch point,” Cranny-Evans said. “So if it doesn’t pick the rocket up from its launch point because it’s not within range, it can’t provide a targeting solution.” US soldiers set up an AN/TPQ-53 Q-53 counter-fire target acquisition radar during an exercise in Hawaii in June. Steph Sweeney In addition, counter-battery radars are set to scan for incoming shells and rockets passing through a specific sector at a specific height. “Unless the radar happens to be looking in the right place at the right time, it will not detect a HIMARS launch,” Cranny-Evans said. “I doubt the Russians have enough counter-battery radars to provide continuous coverage and so would be limited in their ability to provide persistent monitoring even if they could get within range of the rockets.” Counter-battery radars also give an approximate location of the firing battery. While this may be good enough to lay down a general barrage and hope to hit something, it’s not accurate enough for a precise shot. This is especially true for mobile artillery, such as truck-mounted HIMARS and armored self-propelled howitzers, that use “shoot and scoot” tactics to fire a salvo and relocate within minutes. That tactic requires counter-battery fire to be launched within minutes of detecting incoming fire – and Russia’s command structure has been too slow to do this. The HIMARS article reveals the depth of Russian frustration with Ukraine’s new Western-supplied weapons. When Russian forces invaded in February, their more modern artillery outranged Ukraine’s older Soviet-era arms. Now the shoe is on the other foot. The article also suggests that someone in Moscow is looking for scapegoats. Rather than fixing poor equipment or ineffective procedures, they find it easier to blame defeat on enemy secret weapons. Data accessed includes customer names, email addresses, phone numbers and delivery addresses, and in some instances, dates of birth (if provided to purchase alcohol). MyDeal does not store payment, drivers licence or passport details and no customer account passwords or payment details have been compromised. No other Woolworths system was involved in the breach. 2. QLD e-scooter penalties ‘designed to hurt’ Queensland Premier Annastacia Palaszczuk announced Friday afternoon that from November 1, tough penalties will apply for those using an e-scooter in the state. The biggest fine will be $1,078 for using your phone while riding, with drink driving the next highest penalty, at $431. The concerned authorities have described the shortage of staff and resource constraint behind uncollected waste as the situation is getting tougher with every passing day due to population influx as well as proper mechanism of waste segregation and treatment. “We have the capacity to collect only 700 tone waste daily against a daily bulge of 1300 tone,” said Manager Operations MWMC Anwar-ul-Haq. “The company is covering 68 urban Union Councils housing over two million people with only 2200 staff including 1500 sanitary workers.” “Expecting 100 percent results with such staff strength was unjust,” the Manager Operations said, revealing that no recruitment of workforce or acquisition of new machinery was made since 2013. ‘The Hospital Management Rules’ were introduced in 2005 necessitating yellow-bagged waste to be disposed by burning and then dump it at a landfill site or dispose it of through any other method approved by the concerned Federal or Provincial agencies. Lack of professional human resource to collect and properly dump or treat solid waste is yet another reason contributing to environmental degradation and hazards to human health. Chief Executive Officer (CEO) MWMC Muhammad Farooq Dogar said that international standards require a sanitary worker deployed for every 1000 persons for proper sanitation and waste management. “This is missing in our case. To fill the gap of company’s limitations, special campaigns are launched periodically targeting specific areas to cover maximum, if not 100 per cent.” Moreover, he said, there was no practice of solid waste segregation to manage different kinds of waste properly. “Waste segregation would ensure proper management and disposal in accordance with the nature of waste.” Officials are busy these days, in searching a new landfill site and locations including 17-Kassi, Labar Morr and Head Muhammad Wala are in focus. Updated: 17 Oct 2022, 12:52 AM IST Shouvik Das Premium Inflation and geopolitical tensions, among others, are being cited as key factors for depressed demand. istock Experts say inventories have risen to very high levels due to subdued consumer demand NEW DELHI : Chipmakers such as Taiwan Semiconductor Manufacturing Company (TSMC) and Hon Hai Precision Industry Co., better known as Foxconn, may be forced to rethink their investments in the coming quarters as they take a cautious stance to respond to global headwinds such as rising inventory coupled with concerns of rising inflation and geopolitical tensions. TSMC reported on Thursday an 80% jump in net profit for the September quarter at $8.8 billion. However, even as the results beat market estimates, chief executive C.C. Wei said that net capital expenditure for the rest of FY23 has been cut by 10% to $36 billion. He said also that there could be a “likely decline” in the global semiconductor market, and TSMC is “not immune” from the downturn despite its dominant market position. Foxconn, too, advised caution in its outlook for the rest of the year while announcing its September sales. Even as it reported record monthly revenue of $25.9 billion last month, a sequential rise of 83%, the company cited “the dynamics of inflation, the pandemic, and the supply chain” as the key factors of concern. Industry analysts say that the reduced capex plan among global supply chain makers is not unexpected since inventories have risen to very high levels due to subdued consumer demand. Navkendar Singh, associate vice-president of client devices at market researcher International Data Corporation (IDC) India, said the slowdown projection is in line with tapering demand globally. “Even enterprises are spending cautiously, and orders are getting delayed or cancelled, as companies put a freeze on hiring and rethink growth plans,” he said. The demand slowdown has been projected for the immediate quarters in the global information technology (IT) services sectors, too. While Indian software services majors Tata Consultancy Services, Infosys, HCL Technologies and Wipro posted revenue growth, analysts noted a drop in new and active deals among most IT firms indicating a cautious stance among enterprises. Stakeholders in the Indian semiconductor industry are, however, confident that there would not be a direct impact on India’s ability to attract investments in its fledgling semiconductor industry. Vivek Tyagi, chairman of the India Electronics and Semiconductor Association (IESA) said there could be some slowdown in investments by global chipmakers in the coming quarters. “However, it is important to note that semiconductor investments in the country would not come off the back of cutting-edge chips, which is where the slowdown would be. For instance, the automotive sector in India has a demand for chips that are between 28nm (nanometer) and 90nm die size— and this sector is likely to continue with a simmering demand going forward,” he said. Tyagi also said that the Indian semiconductor industry is presently at the stage of attracting investments for chip making factories, which are largely long-term efforts. “As we see it, the demand slowdown and caution in expansion of investments would remain for another 18 to 24 months, following which the global supply chain would stabilize. India could attract assembly, testing, marking and packaging (ATMP) and outsourced semiconductor assembly and test (OSAT) facilities,” he said. The Indian government’s production linked incentive (PLI) plan for semiconductors offers a 50% cash benefit for firms that invest in semiconductor facilities, without a minimum cap on investments. Tyagi said that this, coupled with state-wise land and employment benefits, could see India continuing to receive investments for the semiconductor sector even if it is at a slightly reduced pace.Rajeev Khushu, advisor and board member of IESA, said Bengaluru already has over 80 companies associated with semiconductor manufacturing. None of them were particularly complex, but they featured popular characters and contained addictive gameplay. Many of them have since been removed, though there are plenty of lessons that can be learned from them. Lately, the cruise industry has been spending millions on a multi-medium marketing campaign that includes traditional TV and internet ads as well as placements in reality TV shows. Below, we take a deep dive into the cruise industry’s reputation rehabilitation. The Cruise Industry Wants You Back I’ve never understood the appeal of cruises, as being stuck on a giant barge out at sea, with no land in sight, with the public sounds unpleasant at best and downright scary at worst. Add in a deadly airborne virus that’s easily transmissible between anybody within six feet of one another and you’ve got yourself a prime cocktail for sinking cruise ticket sales. However, recently cruises have been attempting a comeback in a big way, and collaborating with some large brands to do it. The Bachelorette’s latest season featured a cruise as its picturesque backdrop with Virgin Voyages, and the show also gave away a cruise to everyone in their live studio audience for the Men Tell All pre-finale episode. God knows how much this partnership cost the cruise line. The Real Love Boat on CBS is a dating reality show that takes place on a cruise, paying homage to the classic scripted TV show from the 70’s. Disney announced their newest addition to the Disney Cruise Line at their D23 conference earlier this summer, The Disney Treasure, and who can deny the fascinating world of swingers on #CruiseTok? Don’t ask me why pineapples are THE symbol used by those open to getting to know others, intimately, on adventures out at sea, I’m not sure. But if it isn’t entertaining to binge-watch this world from afar. All of this begs the question: is cruising cool now? Or is the industry just trying to make you THINK it’s cool, and really it’s the same cesspool of venereal diseases and bad buffets it always was? We decided to ask an expert who has worked in the industry for decades. virgin cruise ship, are cruises back An Optimistic Outlook on Cruises From an Industry Expert All of this is fine and dandy for some people, but I’m still not convinced a cruise is actually a relaxing vacation with or without COVID-19, and I know I’m not the only one who feels this way. Is the industry optimistic it’ll bounce back as airlines have? We picked the brain of Bob Levinstein, the CEO of CruiseCompete, a cruise booking site that aggregates specials from some of the top cruise lines in the world, to gauge a temperature on the cruise industry as a whole. He explained that CruiseCompete, along with the entire industry, experienced a “major downturn” at the start, and highs and lows throughout the pandemic. “As COVID waves came and went, we were on a roller coaster of hopes and crashes, but we’re finally at the light at the end of the tunnel,” said Levinstein. “Cruise bookings are coming back rapidly, but ships are not yet sailing as full as in a normal year. As a result, there are lots of great deals out there for cruisers right now. Think $50 per person per day for your cabin, all of your meals, and lots of free onboard activities,” said Levinstein. Offering a ton of perks and discounted prices sounds like an industry hungry for customers to come back, we’ll see if it proves to be a lucrative strategy. What’s New in Cruising? I asked Levinstein about all of the cruise-related media spots recently, and whether he sees a renaissance happening within the industry. “Absolutely. Lots of new, innovative ships are starting to sail, with new onboard experiences like roller coasters at sea and new dining and lounge venues,” said Levinstein. “Virgin Voyages offers hip, adults-only sailing. More adventure cruises are offering once-in-a-lifetime experiences in places like Antarctica, the Galapagos, and the Sea of Cortez during whale calving season are capturing travelers’ imaginations.” Okay, even I’ll admit that a cruise to Antarctica sounds amazing, but it’s also one of the only ways to get there if I’m not mistaken. What Should Travelers Know About Cruises and COVID?  I also asked Levinstein about the latest in COVID-19 protections on cruise lines and his messaging was clear: he doesn’t view it as a concern. “Just about all of the COVID-era restrictions are gone. Masks went away months ago. Vaccinated passengers generally have no restrictions at all and unvaccinated passengers can again sail with just a quick test prior to boarding,” said Levinstein. “Even this testing requirement for the unvaccinated has already been dropped by some lines and is likely to go away completely in the next few months.” I also asked him what he would say to folks who are concerned about getting sick on a cruise: “I would say: Think of all of the people you know personally who have had COVID. Ask yourself: how many of those people caught it on a cruise? Zero? Cruises were never the problem; COVID was.” Well there you have it. Cruises are sailing the seven seas once again and it’s up to you whether you’ll indulge in their offerings. Personally, I’ll abstain, although the bumpy return of air travel hasn’t felt like a no-brainer alternative to seasickness during the past few months either. Scientists have now seen these cells in action in lung tissue, as well as other organs that serve as barriers in the body, such as the small intestine, colon and skin. When they used drugs called senolytics to kill these cells, injuries to lung tissues healed more slowly. “Senescent cells can occupy niches with privileged positions as ‘sentinels’ that monitor tissue for injury and respond by stimulating nearby stem cells to grow and initiate repair,” said Tien Peng, MD, associate professor of pulmonary, critical care, allergy and sleep medicine, and senior author of the study, which appears in Science on October 13, 2022. Aging cells can both damage and heal Peng said it was understandable that scientists at first viewed senescent cells as purely detrimental. As people age, senescent cells accumulate that have characteristics of old, worn-out cells, including the inability to make new cells. Instead of dying like normal aged cells, they to live on, spewing a cocktail of inflammatory compounds that form the senescence associated secretory phenotype (SASP). These factors are linked to Alzheimer’s disease, arthritis, and other age-related maladies including cancer. The catchy name “zombie cells” was coined for them. Using senolytics that target and kill “zombie cells,” researchers made the exciting discovery that clearing senescent cells from animals thwarted or diminished age-related disease and extended the lifespan of the animals. Thereafter, a boom of activity ensued in research labs and pharmaceutical companies focused on discovering and refining more powerful versions of these drugs. But killing off senescent cells has dangers, Peng said. For one thing, this current study showed that senescent cells also possess the ability to promote normal healing through activation of stem cell repair. “Our study suggests that senolytics could adversely affect normal repair, but they also have the potential to target diseases where senescent cells drive pathologic stem cell behavior,” said Peng. Lighting up senescent cells One major challenge to studying senescent cells is that biomarkers of senescence (such as the gene p16) are often quite sparse, making it difficult to detect the cells. In early experiments, researchers extracted cells called fibroblasts into culture dishes, allowing them to grow and produce enough cells to experiment with, and then stressed the cells with chemicals that induced them to become senescent. But in living organisms, cells interact with tissues around them, strongly affecting the cells’ gene activity. This means that the characteristics of cells growing isolated in a glass dish could be quite different from that of cells in their natural environment. To create a more powerful tool for their studies, postdoctoral scholar Nabora Reyes de Barboza, Ph.D. and colleagues improved on a common technique of fusing a relevant gene—in this case, the p16 gene, which is overly active in senescent cells—with green fluorescent protein (GFP) as a marker that can reveal the location of the cells under ultraviolet light. By enhancing the quantity and stability of green fluorescent protein in these senescent cells, Reyes greatly amplified the fluorescent signal, finally enabling the researchers to see senescent cells in their natural habitat of living tissues. “Zombies” stimulate stem cells shortly after birth Using this highly sensitive tool, the researchers found that senescent cells exist in young and healthy tissues to a greater extent than previously thought, and actually begin appearing shortly after birth. The scientists also identified specific growth factors that senescent cells secrete to stimulate stem cells to grow and repair tissues. Relevant to aging and tissue injury is the discovery that cells of the immune system such as macrophages and monocytes can activate senescent cells, suggesting that inflammation seen in aged or damaged tissue is a critical modifier of senescent cell activity and regeneration. In their studies of lung tissue, Peng’s team observed green glowing senescent cells lying next to stem cells on the basement membrane that serves as a barrier preventing foreign cells and harmful chemicals from entering the body and also allows oxygen to diffuse from air in the lungs into underlying tissues. Damage can occur at this dynamic interface. Using this highly sensitive tool, the researchers found that senescent cells exist in young and healthy tissues to a greater extent than previously thought, and actually begin appearing shortly after birth. Image is in the public domain The team saw senescent cells in similar positions in other barrier organs such as small intestine, colon, and skin, and their experiments confirmed that if senescent cells were killed with senolytics, lung stem cells were not able to properly repair the barrier surface. Science Abstract Sentinel p16INK4a+ cells in the basement membrane form a reparative niche in the lung We engineered an ultrasensitive reporter of p16INK4a, a biomarker of cellular senescence. Our reporter detected p16INK4a-expressing fibroblasts with certain senescent characteristics that appeared shortly after birth in the basement membrane adjacent to epithelial stem cells in the lung. Furthermore, these p16INK4a+ fibroblasts had enhanced capacity to sense tissue inflammation and respond through their increased secretory capacity to promote epithelial regeneration. In addition, p16INK4a expression was required in fibroblasts to enhance epithelial regeneration. This study highlights a role for p16INK4a+ fibroblasts as tissue-resident sentinels in the stem cell niche that monitor barrier integrity and rapidly respond to inflammation to promote tissue regeneration.
