Endocrinopathies after Allogeneic and Autologous Transplantation of Hematopoietic Stem Cells
Endocrinopathies after Allogeneic and Autologous Transplantation of Hematopoietic Stem Cells
Early and late endocrine disorders are among the most common complications in survivors after hematopoietic allogeneic- (allo-) and autologous- (auto-) stem cell transplant (HSCT). This review summarizes main endocrine disorders reported in literature and observed in our center as consequence of auto- and allo-HSCT and outlines current options for their management. Gonadal impairment has been found early in approximately two-thirds of auto- and allo-HSCT patients: 90–99% of women and 60–90% of men. Dysfunctions of the hypothalamus-pituitary-growth hormone/insulin gro...

Establishment of Etv5 gene knockout mice as a recipient model for spermatogonial stem cell transplantation | Biology Open
Spermatogonial stem cell (SSC) transplantation is an alternative reproductive method to achieve conservation and production of elite animals in livestock production. Creating a recipient animal without endogenous germ cells is important for effective SSC transplantation. However, natural mutants with depletion of SSCs are difficult to obtain, and drug ablation of endogenous germ cells is arduous to perform for practical use. In this study, we used mouse models to study the preparation of recipients with congenital germ cell ablation. We knocked out (KO) Ets-variant gene 5 ( Etv5 ) in mice u...

Evidence for Stem Cell Transplantation as a Cure for HIV-1 - Beyondfitnessforever
A long-term follow-up study of a second London patient has demonstrated a successful stem cell transplantation from donors harboring an HIV-resistant gene. Blood taken from the patient revealed an absence of no active viral infection 30 months following cessation of antiviral therapy. Exploiting Gene Expression Profiling to Identify Novel Minimal Residual Disease Markers of Neuroblastoma | Clinical Cancer Research
Exploiting Gene Expression Profiling to Identify Novel Minimal Residual Disease Markers of Neuroblastoma | Clinical Cancer Research
Purpose: Minimal residual disease (MRD) presents a significant hurdle to curing metastatic neuroblastoma. Biological therapies directed against MRD can improve outcome. Evaluating treatment efficacy requires MRD measurement, which serves as surrogate endpoint. Because of tumor heterogeneity, no single marker will likely be adequate. Genome-wide expression profiling can uncover potential MRD markers differentially expressed in tumors over normal marrow/blood. Experimental Design: Gene expression array was carried out on 48 stage 4 tumors and 9 remission marrows using the Affymetrix U95 gene ...

FDA approves new drug for men at high risk of prostate cancer spread - Harvard Health Blog - Harvard Health Publishing
A newly approved drug called apalutamide is giving hope to thousands of men confronting a tenacious problem after being treated for prostate cancer. Prostate-specific antigen (PSA) levels should plummet to zero after surgery, and to near zero after radiation therapy, but in some men, they continue rising even when there’s no other evidence of cancer […]

Frontiers | Novel Biomarkers for Outcome After Allogeneic Hematopoietic Stem Cell Transplantation | Immunology
Allogeneic hematopoietic stem cell transplantation (allo-HSCT) is a well-established curative treatment for various malignant hematological diseases. However, its clinical success is substantially limited by major complications including graft-vs.-host disease (GVHD) and relapse of the underlying disease. Although these complications are known to lead to significant morbidity and mortality, standardized pathways for risk stratification of patients undergoing allo-HSCT are lacking. Recent advances in the development of diagnostic and prognostic tools have allowed the identification of biomar...

Genotypes of NK Cell KIR Receptors, Their Ligands, and Fcγ Receptors in the Response of Neuroblastoma Patients to Hu14.18-IL2 Immunotherapy | Cancer Research
Response to immunocytokine (IC) therapy is dependent on natural killer cells in murine neuroblastoma (NBL) models. Furthermore, killer immunoglobulin-like receptor (KIR)/KIR-ligand mismatch is associated with improved outcome to autologous stem cell transplant for NBL. Additionally, clinical antitumor response to monoclonal antibodies has been associated with specific polymorphic-Fc γ R alleles. Relapsed/refractory NBL patients received the hu14.18-IL2 IC (humanized anti-GD2 monoclonal antibody linked to human IL2) in a Children's Oncology Group phase II trial. In this report, these patient...

Granulin–Epithelin Precursor Renders Hepatocellular Carcinoma Cells Resistant to Natural Killer Cytotoxicity | Cancer Immunology Research
Granulin–Epithelin Precursor Renders Hepatocellular Carcinoma Cells Resistant to Natural Killer Cytotoxicity | Cancer Immunology Research
Immunoevasion is an emerging hallmark of cancer. Impairment of natural killer (NK) cytotoxicity is a mechanism to evade host immunosurveillance. Granulin–epithelin precursor (GEP) is a hepatic oncofetal protein regulating growth, invasion, and chemoresistance in hepatocellular carcinoma (HCC). We examined the role of GEP in conferring HCC cells the ability to evade NK cytotoxicity. In HCC cell lines, GEP overexpression reduced, whereas GEP suppression enhanced sensitivity to NK cytotoxicity. GEP downregulated surface expression of MHC class I chain–related molecule A (MICA), ligand for NK s...

High-fidelity and time-driven simulation of large wireless networks with parallel processing - IEEE Journals & Magazine
Parallel processing is a promising technique for reducing the execution time of a large and complex wireless network simulation. In this article, a parallel processing technique is presented for time-driven simulations of large and complex wireless networks. The technique explicitly considers the physical-layer details of wireless network simulators such as shadowing and co-channel interference. The technique uses multiple processors interconnected with highspeed links. Principles and issues are elaborated, including workload partitioning, synchronization, and database design. Numerical res...

Human herpes 6 virus encephalitis complicating allogeneic hematopoietic stem cell transplantation | Neurology
Objective: To describe the presentation and management of encephalitis due to human herpes 6 virus (HHV-6) in patients who underwent allogeneic hematopoietic stem cell transplant (alloHSCT), via retrospective chart review. Methods: Of the 243 patients who underwent alloHSCT at the NIH Clinical Center during 2009 to 2011, we retrospectively analyzed 9 diagnosed with HHV-6 encephalitis post-alloHSCT. Results: Eight men and 1 woman (aged 19–60 years) met diagnostic criteria for study inclusion. The median time from HSCT to initial symptoms was 21 days. All patients presented with altered menta...
