Flagellum and toxin phase variation impacts intestinal colonization and disease development in a mouse model of Clostridioides difficile infection
Flagellum and toxin phase variation impacts intestinal colonization and disease development in a mouse model of Clostridioides difficile infection
Clostridioides difficile is a major nosocomial pathogen that can cause severe, toxin-mediated diarrhea and pseudomembranous colitis. Recent work has shown that C. difficile exhibits heterogeneity in swimming motility and toxin production in vitro through phase variation by site- …

Assembly of the exosporium layer in Clostridioides difficile spores
Clostridioides difficile is a Gram-positive, spore-forming obligate anaerobe and a major threat to the healthcare system world-wide. Because of its strict anaerobic requirements, the infectious and transmissible morphotype is the dormant spore. During infection, C. difficile produces spores that can …

Glucosyltransferase-dependent and independent effects of Clostridioides difficile toxins during infection
Clostridioides difficile infection (CDI) is the leading cause of nosocomial diarrhea and pseudomembranous colitis in the USA. In addition to these symptoms, patients with CDI can develop severe inflammation and tissue damage, resulting in life-threatening toxic megacolon. CDI is mediated by two larg …

Identification of novel, cryptic Clostridioides species isolates from environmental samples collected from diverse geographical locations
Clostridioides difficile is a pathogen often associated with hospital-acquired infection or antimicrobial-induced disease; however, increasing evidence indicates infections can result from community or environmental sources. Most genomic sequencing of C. difficile has focused on clinic …

Crosstalk Between the Gut Microbiota and Epithelial Cells Under Physiological and Infectious Conditions
The gastrointestinal tract (GIT) is considered the largest immunological organ, with a diverse gut microbiota, that contributes to combatting pathogens and maintaining human health. Under physiological conditions, the crosstalk between gut microbiota and intestinal epithelial cells (IECs) plays a cr …

Multiple Regulatory Mechanisms Control the Production of CmrRST, an Atypical Signal Transduction System in Clostridioides difficile
Clostridioides difficile, an intestinal pathogen and leading cause of nosocomial infection, exhibits extensive phenotypic heterogeneity through phase variation. The signal transduction system CmrRST, which encodes two response regulators (CmrR and CmrT) and a sensor kinase (CmrS), impacts C. diffici …

Binary Toxin Expression by Clostridioides difficile Is Associated With Worse Disease
Patients with fecal samples that were positive for CDT had increased disease severity and worse clinical outcomes. Utilization of PCR and testing for C. difficile toxins A and B may not reveal the entire picture when diagnosing CDI; detection of CDT-expressing strains is valuable in identifyi …

Seeing C diff differently - The Lancet
Seeing C diff differently - The Lancet
The first time I had a Clostridioides difficile infection, I was 37 years old. After a sequential antibiotic cocktail of amoxicillin, penicillin, metronidazole, and clindamycin prescribed to combat an infection in my jaw, I had the first wave of symptoms. Even having a long history of stomach-related issues dating back to my childhood, these new symptoms were like nothing I had ever experienced. Anything I consumed quickly traversed my gastrointestinal system within an impressively short timeframe, leaving me with a feeling of perpetual emptiness and exhaustion, a heightened awareness of the nearest restroom facilities, and an existential appreciation for saltine crackers.
