Objective: The protective effect of aloe vera is due to its anti-inflammatory properties. This experimental study focused on revealing the effects of aloe vera on antioxidant systems and determining the number of degenerated neurons after cerebral ischemia and reperfusion. Methods: Twenty-one rats were divided equally into three groups: group I rats were used as controls; group II rats were fed with standard diet, and group III rats were fed with standard diet plus aloe vera for 30 days. For groups II and III, the right middle cerebral arteries were occluded for 45 min and then reperfused for 24 h. After these procedures, rats in all groups were sacrificed. The levels of malondialdehyde and nuclear respiratory factor-1 and superoxide dismutase activity in the right cerebral cortex were measured. The number of degenerated neurons was counted in histological samples of the right cerebral cortex. Neuronal nitric oxide synthase expression was examined with immunohistochemical methods. Background Aspirin is one of the most popular NSAIDs worldwide because of its anti-inflammatory and anticoagulant effects, and however, gastrointestinal injury remains a major complication. We previously reported co-lyophilized aspirin/trehalose (Lyo A/T) decreased the aspirin-induced gastric lesions in dogs. Aim This study investigated the mechanism of gastroprotective effects of trehalose in vitro and in vivo. Methods The apoptotic assays were performed in a human gastric carcinoma cell line, which was treated with aspirin, mixed aspirin/trehalose (Mix A/T) or Lyo A/T. Gastric ulcer severity was examined after oral administration of drugs in rats. In addition, the mucosal tissue apoptotic status in drug-treated rats was evaluated. Molecular dynamics simulations and laser Raman spectroscopy were performed in order to examine the molecular properties of Lyo A/T. Results DNA fragmentation was detected in AGS cells that were treated with aspirin and Mix A/T, but not in the Lyo A/T-treated cells. There were fewer apoptotic cells in the Lyo A/T-treated cells than in the other cells. Gastric injury was reduced in rats that received oral Lyo A/T compared with the others, while PGE2 synthesis was equally decreased in all groups. TUNEL assay and immunohistochemistry of cleaved caspase-3 in the mucosal tissues also revealed that Lyo A/T treatment induced less apoptosis than the others. The Lyo A/T spectrum showed clear differences in several Raman bands compared with that of Mix A/T. Conclusions Our data showed that co-lyophilization of aspirin with trehalose reduced gastric injury, potentially through suppression of aspirin-induced mucosal cell apoptosis while retaining its anti-inflammatory effects.

A plethora of publications have reported several cytotoxic effects associated with chronic consumption of thermoxidized palm oil. This research aims to investigate the effects of garlic and Aloe vera on liver function and hepatic cytoarchitecture in rats fed thermoxidized palm oil diet.,Thirty-five male albino Wistar rats weighing 150-180 g were used for this study. They were randomly assigned into five groups (n = 7): control, thermoxidized palm oil diet fed (TPO), TPO plus garlic juice (TPO + G), TPO plus Aloe gel (TPO + A) and TPO plus garlic/Aloe gel (TPO + G + A). The TPO diet was prepared by mixing 15 g of cooled thermoxidized palm oil with 85 g of rat feed. The juice and gel were orally administered at doses of 2.00 ml/kg and 19.12 ml/kg, respectively. After 3 months of feeding and administration, the animals were sacrificed using standard methods and blood collected via cardiac puncture for analysis.,Serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase were significantly increased (p < 0.001) in the TPO group compared to the control. This increase was reduced (p < 0.001) in all the treated groups compared to TPO, with the greatest decrease (p < 0.001) seen in TPO + A. Total bilirubin was increased (p < 0.001) in the TPO group compared to the control, whereas there was a significant decrease (p < 0.001) in all the treated groups. Serum proteins and plasma fibrinogen were lowered (p < 0.001) in the TPO group compared to the control but increased progressively in all the treated groups. TPO induced prominent histopathological derangements of the liver tissues. However, there were marked improvements following treatment with garlic and Aloe vera.,The results obtained in this study have revealed that chronic consumption of thermoxidized palm oil is hazardous to health by inducing hepatotoxicity, as seen in increased ALT, AST and total and unconjugated bilirubin and decreased total protein. However, these debilitating effects were seen to be greatly ameliorated following garlic juice and Aloe vera gel administration. If these results are to be extrapolated to humans, then the chronic consumption of thermoxidized palm oil diet should be seriously discouraged.

Biomolecules Free Full-Text Trehalose for Ocular Surface Health
Trehalose is a natural disaccharide synthesized in various life forms, but not found in vertebrates. An increasing body of evidence demonstrates exceptional bioprotective characteristics of trehalose. This review discusses the scientific findings on potential functions of trehalose in oxidative stress, protein clearance, and inflammation, with an emphasis on animal models and clinical trials in ophthalmology. The main objective is to help understand the beneficial effects of trehalose in clinical trials and practice, especially in patients suffering from ocular surface disease. The discussion is supplemented with an overview of patents for the use of trehalose in dry eye and with prospects for the 2020s.

Background: Applying a cooling treatment from outside the body is one way to lower fever in children. Midwives commonly perform a warm water compress in their care, but it is considered less effective. Thus, compress with Aloe Vera Barbadensis Miller is proposed in this study as an alternative treatment. Objective: To examine the effect of Aloe Vera Barbadensis Miller compress in reducing body temperature in children with fever. Methods: This was a quasi-experimental study with pretest and posttest with control group design. Forty children were recruited, with twenty of them assigned in each group. Purposive sampling was performed to select the sample. A digital rectal thermometer was used to measure the temperature. Independent t- test and paired test were used for data analysis. Results: Findings showed that the Aloe Vera compress group showed a higher decrease of body temperature compared to the warm water compress group. The difference of body temperature after 20 minutes in the experiment group was 1.435 while in the control group was only 1.085. There were statistically significant differences in body temperature between the experiment and control group (p=0.013). Conclusion: Aloe Vera Barbadensis Miller compress is an effective alternative therapy in reducing body temperature in children with fever.  It is recommended for midwives to apply this intervention to reduce body temperature significantly.

Background: Intensive care units (ICUs) admissions due to sepsis represent 2 to 11%. Although there are well stablished&nbsp; therapeutic available, it still is the major cause of death in most ICUs and the leading cause of acute kidney injury. The objective of this work was to evaluate the role of Aloe vera extract on survival rates and kidney histopathology of mice submitted to sepsis induced by cecal ligation and puncture. Methods and Findings: Twenty six mice were distributed into four groups: Sham - surgery simulation (N=5); Control - Cecal ligation and puncture was performed (N=7); Aloe - Surgery simulation and treatment with Aloe vera extract (N=7); and Sepsis + Aloe - Cecal ligation and puncture was performed followed by treatment with Aloe vera extract (N=7). The animals were observed until death. Kidneys were analysed by histopathology. Kruskall-Wallis test was used to compare the histopathological results. Survival analysis was assessed by Kaplan-Meier statistics and compared using log rank test. A significance level of 5% was adopted. The survival time ranged from 19 hours for the Control group up to 24 hours for the animals of Sepsis + Aloe group, corresponding to a survival time of 1.26 times greater (p0.05). Conclusions: Aloe vera extracts delays mortality but does not attenuate kidney injury in sepsis induced by cecal ligation and puncture in mice.

It is well established that trehalose (TRH) affects the physical properties of lipid bilayers and stabilizes biological membranes. We present molecular dynamics (MD) computer simulations to investigate the interactions between lipid membranes formed by 1,2-dimyristoyl-sn-glycero-3-phosphocholine (DMPC) and TRH. Both atomistic and coarse-grained (CG) interaction models were employed, and the coarse graining of DMPC leads to a reduction in the acyl chain length corresponding to a 1,2-dilauroyl-sn-glycero-3-phosphocholine lipid (DLPC). Several modifications of the Martini interaction model, used for CG simulations, were implemented, resulting in different potentials of mean force (PMFs) for DMPC bilayer–TRH interactions. These PMFs were subsequently used in a simple two-site analytical model for the description of sugar binding at the membrane interface. In contrast to that in atomistic MD simulations, the binding in the CG model was not in agreement with the two-site model. Our interpretation is that the interaction balance, involving water, TRH, and lipids, in the CG systems needs further tuning of the force-field parameters. The area per lipid is only weakly affected by TRH concentration, whereas the compressibility modulus related to the fluctuations of the membrane increases with an increase in TRH content. In agreement with experimental findings, the bending modulus is not affected by the inclusion of TRH. The important aspects of lipid bilayer interactions with biomolecules are membrane curvature generation and sensing. In the present investigation, membrane curvature is generated by artificial buckling of the bilayer in one dimension. It turns out that TRH prefers the regions with the highest curvature, which enables the most favorable situation for lipid–sugar interactions.

Trehalose is a nonreducing disaccharide that has recently attracted much attention because of its ability to inhibit protein aggregation, induce autophagy, and protect against dissections and strokes. In vertebrates, the biosynthesis of trehalose was long considered absent due to the lack of annotated genes involved in this process. In contrast, trehalase (TreH), which is an enzyme required for the cleavage of trehalose, is known to be conserved and expressed. Here, we show that trehalose is present as an endogenous metabolite in the rodent hippocampus. We found that primary astrocytes were able to synthesize trehalose and release it into the extracellular space. Notably, the TreH enzyme was observed only in the soma of neurons, which are the exclusive users of this substrate. A statistical analysis of the metabolome during different stages of maturation indicated that this metabolite is implicated in neuronal maturation. A morphological analysis of primary neurons confirmed that trehalose is required for neuronal arborization.
