Evaluation of the Nutritional and Metabolic Effects of Aloe vera - Herbal Medicine - NCBI Bookshelf
Aloe vera has a long history of popular and traditional use. It is used in traditional Indian medicine for constipation, colic, skin diseases, worm infestation, and infections (Heber 2007). It is also used in Trinidad and Tobago for hypertension (Lans 2006) and among Mexican Americans for the treatment of type 2 diabetes mellitus (DM; Coronado et al. 2004). In Chinese medicine, it is often recommended in the treatment of fungal diseases (Heber 2007). In Western society, Aloe vera is one of the few herbal medicines in common usage, and it has found widespread use in the cosmetic, pharmaceutical, and food industries. In the case of health, the therapeutic claims for the topical and oral application of Aloe vera cover a wide range of conditions, but few claims have been the subject of robust clinical investigation. The conditions for which clinical trials of Aloe vera have been conducted include skin conditions, management of burn and wound healing, constipation, DM, and gastrointestinal disorders.

Toxics | Free Full-Text | Amelioration of Hepatotoxic and Neurotoxic Effect of Cartap by Aloe vera in Wistar Rats
Pesticide exposure can pose a serious risk to nontarget animals. Cartap is being broadly used in agricultural fields. The toxic effects of cartap on the levels of hepatotoxicity and neurotoxicity have not been properly studied in mammalian systems. Therefore, the present work focused on the effect of cartap on the liver and brain of Wistar rats and made an assessment of the ameliorating potential of A. vera. The experimental animals were divided into 4 groups, comprising six rats in each: Group 1—Control; Group 2—A. vera; Group 3—Cartap; and Group 4—A. vera + Cartap. The animals orally given cartap and A. vera were sacrificed after 24 h of the final treatment and histological and biochemical investigations were conducted in liver and brain of Wistar rats. Cartap at sublethal concentrations caused substantial decreases in CAT, SOD, and GST levels in the experimental rats. The activity levels of transaminases and phosphatases in cartap group were also found to be substantially altered. The AChE activity was recorded as decreasing in RBC membrane and brain of the cartap-treated animals. The TNF-α and IL-6 level in serum were increased expressively in the cartap challenged groups. Histological investigation of liver showed disorganized hepatic cords and severely congested central veins due to cartap. However, the A. vera extract was observed to significantly protect against the effects of cartap toxicity. The protective impact of A. vera against cartap toxicity may be due to the existence of antioxidants in it. These findings suggest that A. vera may be developed as a potential supplement to the appropriate medication in the treatment of cartap toxicity.

The Protective Effects of Aloe Vera Gel Extract on the Sexual Tissue Damage Caused by Diabetes in Male Rats
Introduction: Diabetics usually suffer from chronic complications, including microvascular and macrovascular disorders. The capillaries supplying the sexual organs affect normal sexual functions. The erectile process has been shown to be impaired in diabetics. Vascular damage in diabetics may be associated with decreased testosterone production, sperm count, testicular weight, and seminiferous tubule diameter. In this study, we investigated the effects of Aloe vera gel extract (from Sistan and Baluchistan in Iran) on male genital in streptozocin-induced diabetic rats. Methods: Of twenty-four male Wistar rats, diabetes was induced in 12 rats by intraperitoneal injection of a single dose of STZ. The rats were divided into four groups. The first and third groups received saline (NaCl) daily by gavage. The second and fourth groups received aloe vera gel extract daily by gavage. The treatments were continued for 30 days. At the end of the treatment period, blood samples were collected and serum glucose and testosterone levels were measured by photometric methods. Histological examinations were performed on the prostate and testes. Results: Mean weight index, serum levels of glucose and testosterone, mean sperm count, germline thickness, and seminiferous tubule diameter improved significantly in the diabetes +Aloe vera group. Conclusion: We concluded that consumption of Aloe vera gel extract improved sexual complications in type 1 diabetic rats.

A Mini-Review Possible Mechanisms of Hepatoprotective Effect of Aloe Vera Gel Handayani Indonesian Journal of Cancer Chemoprevention
Protective agent for hepatotoxicity is still a great challenge in the management of liver diseases. Aloe vera is a beneficial plant that has been studied for food supplements, cosmetic and herbal medicine. Aloe vera contains many compounds which have a role in body health including polysaccharides, phenolic, flavonoid, terpenoid, amino acid, and several minerals. There have been compelling evidences that natural phytochemicals and their derivatives have hepatoprotective activities. Information of the aloe vera and its mechanism of action for possible hepatoprotective activities, including in silico, in vitro, and in vivo studies were obtained from Pubmed, Science Direct, Scopus, and Google scholar search engines. This current review was focusing on the possible contribution of compounds inside aloe vera gel and the suggestion of its mechanism on protective effect, especially for liver. The complexity of monosaccharides composition, backbone structures, acetyl group, and molecular weight of aloe polysaccharides have possible correlations with its hepatoprotective effect. Most of the hepatoprotective mechanisms of aloe compounds are related to their protective effect against inflammation and oxidative stress. Several compounds may have combination effects or several targets lead to synergistic effects. Keywords: Aloe vera, food supplement, hepatoprotective, liver disease, mechanism of action.
Foods Free Full-Text Dry but Not Humid Thermal Processing of Aloe vera Gel Promotes Cytotoxicity on Human Intestinal Cells HT-29
Aloe vera products, both in food and cosmetics, are becoming increasingly popular due to their claimed beneficial effects, which are mainly attributed to the active compound acemannan. Usually, these end products are based on powdered starting materials. High temperatures during the drying process to obtain the starting materials have several advantages, like shortening the drying time, eliminating toxic aloin and reducing bacterial contamination. Nevertheless, there are two major drawbacks: first, at temperatures of 80 °C or higher, structural changes in acemannan, especially its deacetylation (>46%), are triggered, which does not happen at lower temperatures (14% at 60 °C); secondly, a toxic principle is formed at higher temperatures, resulting in a higher cytotoxicity. Thus, two temperature-dependent but opposing effects cause with a median cytotoxic concentration of CC50 = 0.4× a peak of cytotoxicity at 80 °C; at 60 °C this cytotoxic substance is not formed and at 100 °C aloin is more readily eliminated, resulting in a CC50 = 1.1× and CC50 = 1.4×, respectively. The cytotoxic substance generated by dry heat at 80 °C is not a modified polysaccharide because its polysaccharide-enriched alcohol-insoluble fraction is with CC50 = 0.9× less cytotoxic. Moreover, this substance is polar enough to be washed away with ethanol. Additionally, when Aloe gel is heated at 80 °C under humid conditions (pasteurization), the cytotoxicity does not increase (CC50 = 1.6×). Finally, to produce powdered starting materials from Aloe gel, it is recommended to use temperatures of around 60 °C in order to preserve the acemannan structure (and thus biological activity) and the low cytotoxicity.
