Antioxidants Modulation of Hydrogen Peroxide Cytotoxicity by Antioxidant Aloe vera Extract
Although Aloe vera contains numerous bioactive components, the activity principles of widely used A. vera extracts are uncertain. Therefore, we analyzed the effects of genuine A. vera aqueous extract (AV) on human cells with respect to the effects of hydrogen peroxide (H2O2) and 4-hydroxynonenal (HNE). Fully developed A. vera leaves were harvested and analyzed for vitamin C, carotenoids, total soluble phenolic content, and antioxidant capacity. Furthermore, human cervical cancer (HeLa), human microvascular endothelial cells (HMEC), human keratinocytes (HaCat), and human osteosarcoma (HOS) cell cultures were treated with AV extract for one hour after treatment with H2O2 or HNE. The cell number and viability were determined using Trypan Blue, and endogenous reactive oxygen species (ROS) production was determined by fluorescence, while intracellular HNE–protein adducts were measured for the first time ever by genuine cell-based HNE–His ELISA. The AV extract expressed strong antioxidant capacities (1.1 mmol of Trolox eq/g fresh weight) and cell-type-specific influence on the cytotoxicity of H2O2, as well as on endogenous production of ROS and HNE–protein adducts induced by HNE treatment, while AV itself did not induce production of ROS or HNE–protein adducts at all. This study, for the first time, revealed the importance of HNE for the activity principles of AV. Since HMEC cells were the most sensitive to AV, the effects of AV on microvascular endothelia could be of particular importance for the activity principles of Aloe vera extracts.

The nutraceutical properties of Aloe vera have been attributed to a glucomannan known as acemannan. Recently information has been published about the presence of fructans in Aloe vera but there are no publications about acemannan and fructans as prebiotic compounds. This study investigated in vitro the prebiotic properties of these polysaccharides. Our results demonstrated that fructans from Aloe vera induced bacterial growth better than inulin (commercial FOS). Acemannan stimulated bacterial growth less than fructans, and as much as commercial FOS. Using qPCR to study the bacterial population of human feces fermented in a bioreactor simulating colon conditions, we found that fructans induce an increase in the population of Bifidobacterium spp. Fructans produced greater amounts of short chain fatty acids (SCFA), while the branched-chain fatty acids (BCFA) did not increase with these polysaccharides. Acemannan increased significantly acetate concentrations. Therefore, both Aloe vera polysaccharides have prebiotic potentials.

This paper investigates the performance, combustion and emission characteristics of a direct injection (DI) diesel engine, fueled with alumina ($$\hbox {Al}_{2} \hbox {O} _{3})$$ Al2O3) nanoparticles-dispersed aloe vera diesel blends. The test fuels B10 (90% diesel and 10% aloe vera biodiesel), $$\hbox {B10}+ \hbox {Al}_{2} \hbox {O}_{3}$$ B10+Al2O3 (90% diesel, 10% aloe vera biodiesel and 30 ppm of $$\hbox {Al}_{2} \hbox {O}_{3}$$ Al2O3 nanoparticles), B20 (80% diesel and 20% aloe vera biodiesel) and $$\hbox {B20}+ \hbox {Al}_{2} \hbox {O}_{3}$$ B20+Al2O3 (80% diesel, 20% aloe vera biodiesel and 30 ppm of $$\hbox {Al}_{2} \hbox {O}_{3}$$ Al2O3 nanoparticles) are prepared separately for each aloe vera diesel blends at a dosage of 30 ppm with an aid of an ultrasonicator. The combustion characteristics such as heat release rate and cylinder pressure are investigated using the data acquisition system, while the engine performance features are examined by brake-specific fuel consumption and brake thermal efficiency. The levels of engine pollutants are analyzed by unburned hydrocarbon (UBHC), carbon monoxide (CO), nitric oxide (NO) and smoke opacity. These investigations disclosed the use of biodiesel blends with $$\hbox {Al}_{2} \hbox {O}_{3}$$ Al2O3 nanoparticles in DI diesel engine with significant amelioration of combustion characteristic and substantial decrement of CO, NO, smoke, UBHC emissions and brake thermal efficiency by 44%, 47%, 37%, 29% and 3%, when compared with the diesel fuel.

The aim of the study was to determine the anti tumour and radio protective activity of Aloe vera. The tumour study was conducted in ascites tumour model and in solid tumour model. In solid tumour model a significant result was obtained when the extract of Aloe vera was administered to animal models. In radio protective study even though there was no significant effect of drug along with radiation induced animal's body weight, haemoglobin and in differential count there was a slight increase in Total WBC cou nt in drug induced animals along with radiation when compared with radiation alone treated animals

Many studies have proved that bioactive components of Aloe vera have an anti-inflammatory effect and support lipid and carbohydrate metabolism, helping to maintain normal sugar and cholesterol levels in blood and normal body weight. When aloe is applied externally, it accelerates the regeneration of the damaged skin. Aloe contains antioxidants, which may increase the shelf-life and nutritional value of food; therefore, it is widely used in cosmetic, pharmaceutical and food industry. An antioxidant activity was shown for leaf’s skin, flowers and gel of aloe. In this work the future of A. vera as effective antioxidants is primarily discussed and expected trends are summarised. Furthermore, the bioactive components and the health-promoting effects of A. vera are investigated.

Background Aloe’s reported bioactivities (anticancer, anti-inflammatory and wound healing) suggest they might inhibit a subgroup of matrix metalloproteinases (MMPs) called gelatinases (MMP-2 and MMP-9). The goal of the present study was to compare the MMP inhibitory potential of two Aloe species, A. vera and A. arborescens. Methods Different types of extraction were tested and specific bioactive compounds were quantified. Cancer cell invasion inhibitory activities were measured in vitro using the wound healing assay in human colon cancer cells (HT29). Effects on gelatinase activities were further assessed by dye-quenched gelatin and gelatin zymography. Results Different types of extraction yielded significantly different levels of bioactivities and of bioactive compounds, which might be due to a greater amount of extractable bioactive compounds such as anthraquinones. Both A. arborescens and A. vera have potential as inhibitory agents in cancer cell proliferation via MMP-9 and MMP-2 enzymatic activity inhibition, being able to reduce colon cancer cell proliferation and migration but A. arborescens showed to be a more effective inhibitor of cancer cell migration than A. vera. Conclusion This work opens novel perspectives on the mode of action of Aloe species in cancer cell migration and may provide clues as to why there are so many conflicting results on Aloe’s activities.

AIM: Diet is a natural source of butyrate through the fermentation of non-digestive fiber, such as acemannan in Aloe vera gel, is a strong appearing target for health and quality of life as an immune modulation and colorectal cancer prevention in aged people. In our earlier research on fermentation by endophytic bacteria in Aloe vera gel, butyric acid was identified by GC/MSD analysis. Present investigation aims the identification of the microbiota. MATERIALS AND METHODS: The endophytic microbiota of Aloe vera gel in the fermented media were examined by use of matrix-assisted laser desorption ionization-time of flight mass spectrometry. RESULTS: The following microbiota were identified: Bacillus cereus, B. licheniformis, Lactobacillus paralimentarium, Yeast: Clavispora lusitaniae. The safety pattern of the prepared Aloe vera gel was tested on normal non-cancerous cells and indicated the absence of any significant possible toxicity on the cells. Also, the extracted gels showed abilities to regulate the inflammatory responses in the inflammation cell models via the reduction in the amount of induced reactive oxygen species and both COX 1 and 2 enzymes. DISCUSSIOIN: Identification of butyrate-producing endophytic microbiota in Aloe vera gel fermentation and finding of inflamatory as well as antioxidant activities of butyrate in the fermented gel may help explain the known beneficial effects of butyrate in intestinal colon and on colitis. An innovative concept of symbiotics: a combination of Aloe vera gel juice and microbiota: Bacillus cereus, B.licheniformis. Lactobacillus paralimentarium and Clavispora lusitaniae, is a perspective on alleviation of cancer disease and improvement of gastrointestinal health by butyrate fermentation.

Objective: This study was designed to compare the effect of Aloe vera gel with aspirin and celecoxib on platelet aggregation. Study Design: Comparative Study. Setting: Post graduate Medical Institute Lahore, Children Hospital, Lahore. Period: September 2015 to September 2016. Material & Methods: Blood was withdrawn from anti-cubital vein, complete blood count was checked, platelet rich plasma was prepared by centrifuging citrated whole blood and then incubated with  Aloe vera low (AVL), Aloe vera high (AVH), aspirin and celecoxib for 30 minutes at 37C. After adding the agonist arachidonic acid, reading was then taken for 3 minutes and percentage aggregation was recorded. Results: Platelet aggregation with aspirin, AVH and AVL was statistically significantly lower as compared to control and celecoxib groups. Conclusion: This study has demonstrateda dose dependentanti-platelet effect of Aloe vera gel which is comparable to aspirin.

Objective: The protective effect of aloe vera is due to its anti-inflammatory properties. This experimental study focused on revealing the effects of aloe vera on antioxidant systems and determining the number of degenerated neurons after cerebral ischemia and reperfusion. Methods: Twenty-one rats were divided equally into three groups: group I rats were used as controls; group II rats were fed with standard diet, and group III rats were fed with standard diet plus aloe vera for 30 days. For groups II and III, the right middle cerebral arteries were occluded for 45 min and then reperfused for 24 h. After these procedures, rats in all groups were sacrificed. The levels of malondialdehyde and nuclear respiratory factor-1 and superoxide dismutase activity in the right cerebral cortex were measured. The number of degenerated neurons was counted in histological samples of the right cerebral cortex. Neuronal nitric oxide synthase expression was examined with immunohistochemical methods.
