Evaluation of the Treatment Effect of Aloe vera Fermentation in Burn Injury Healing Using a Rat Model
Burn injury is a growing medical problem associated with public health, and few effective agents are available for treatment of this disease. In the present study, a burn injury rat model was developed and the accelerated effect of Aloe vera fermentation on burn injury healing was evaluated. Our results indicated that Aloe vera fermentation could markedly reduce the DPPH (56.12%), O2⋅− (93.5%), ⋅OH (76.12%), Fe2+ chelation (82%), and oxygen-reduction activity (0.28 μg/ml) and significantly inhibited the growth of pathogens S. typhimurium ATCC 13311 (inhibition zone diameter: 14 mm), S. enteritidis ATCC13076 (IZD: 13 mm), S. flexneri ATCC 12022 (IZD: 18 mm), E. coli 44102 (IZD: 10 mm), L. monocytogenes ATCC 19111 (IZD: 18 mm), S. dysenteriae 301 (IZD: 20 mm), S. aureus COWAN1 (IZD: 19 mm), and P. acnes ATCC 11827 (IZD: 25 mm) in vitro. The in vivo results indicated that Aloe vera fermentation produced more eosinophils and fibroblasts and less vessel proliferation compared with the model group on the 14th day, which had greatly accelerated burn injury healing via shedding of the scab and promoting hair growth. ELISA results indicated that Aloe vera fermentation had significantly reduced the production of proinflammatory factors TNF-α and IL-1β () and greatly enhanced the yield of anti-inflammatory factor IL-4 in animal serum (). In addition, the high-throughput sequencing results indicated that Aloe vera fermentation obviously increased the percentage of Firmicutes (65.86% vs. 49.76%), while reducing the number of Bacteroidetes (27.60% vs. 45.15%) compared with the M group at the phylum level. At the genus level, Aloe vera fermentation increased the probiotic bacteria Lactobacillus (3.13% vs. 2.09%) and reduced the pathogens Prevotella (10.60% vs.18.24%) and Blautia (2.91% vs. 16.41%) compared with the M group. Therefore, we concluded that the use of Aloe vera fermentation significantly accelerates burn injury healing via reduction of the severity of inflammation and through modification of gut microbiota.

A plethora of publications have reported several cytotoxic effects associated with chronic consumption of thermoxidized palm oil. This research aims to investigate the effects of garlic and Aloe vera on liver function and hepatic cytoarchitecture in rats fed thermoxidized palm oil diet.,Thirty-five male albino Wistar rats weighing 150-180 g were used for this study. They were randomly assigned into five groups (n = 7): control, thermoxidized palm oil diet fed (TPO), TPO plus garlic juice (TPO + G), TPO plus Aloe gel (TPO + A) and TPO plus garlic/Aloe gel (TPO + G + A). The TPO diet was prepared by mixing 15 g of cooled thermoxidized palm oil with 85 g of rat feed. The juice and gel were orally administered at doses of 2.00 ml/kg and 19.12 ml/kg, respectively. After 3 months of feeding and administration, the animals were sacrificed using standard methods and blood collected via cardiac puncture for analysis.,Serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase were significantly increased (p < 0.001) in the TPO group compared to the control. This increase was reduced (p < 0.001) in all the treated groups compared to TPO, with the greatest decrease (p < 0.001) seen in TPO + A. Total bilirubin was increased (p < 0.001) in the TPO group compared to the control, whereas there was a significant decrease (p < 0.001) in all the treated groups. Serum proteins and plasma fibrinogen were lowered (p < 0.001) in the TPO group compared to the control but increased progressively in all the treated groups. TPO induced prominent histopathological derangements of the liver tissues. However, there were marked improvements following treatment with garlic and Aloe vera.,The results obtained in this study have revealed that chronic consumption of thermoxidized palm oil is hazardous to health by inducing hepatotoxicity, as seen in increased ALT, AST and total and unconjugated bilirubin and decreased total protein. However, these debilitating effects were seen to be greatly ameliorated following garlic juice and Aloe vera gel administration. If these results are to be extrapolated to humans, then the chronic consumption of thermoxidized palm oil diet should be seriously discouraged.

Aloe vera gel is a potential material as raw material industry, this is because a very complex composition. However Aloe vera gel is very easily oxidized or unstable. Viscosity gel and the benefit are decreased at room temperature after 24-36 hours. This research aims to obtain information about the resistance to oxidation via nitogren gas treatment and antioxidants, as well as the influence of phase changes in an attempt to retain the characteristics of the physicochemical Aloe vera gel over time. This Study can be described a conclusion that the best storage conditions are sound-proofed temperature conditions (4 ± 1)oc. Environmental conditioning by administering nitrogen gas storage and antioxidant Buthylated Hydroxytoluene (BHT) 750 ppm for 4 weeks defending the nature physicochemical Aloe vera gel. Based on activation of macrophages and vaccine adjuvant activity by aloe polysaccharide or acemannan, we showed that the importance of butyrate and host-micRNA-microbiota interaction in regulation of gut immunity, and butyrate is a chemical factor capable of facilitating induced pluripotent stem cell generation, an important step for the establishment of pluripotency. Furthermore we exhibited three case reports on the beneficial roles of aloe vera juice-successive ingestion as nutritional and medicinal uses to influenza virus and norovirus.

Background: Intensive care units (ICUs) admissions due to sepsis represent 2 to 11%. Although there are well stablished&nbsp; therapeutic available, it still is the major cause of death in most ICUs and the leading cause of acute kidney injury. The objective of this work was to evaluate the role of Aloe vera extract on survival rates and kidney histopathology of mice submitted to sepsis induced by cecal ligation and puncture. Methods and Findings: Twenty six mice were distributed into four groups: Sham - surgery simulation (N=5); Control - Cecal ligation and puncture was performed (N=7); Aloe - Surgery simulation and treatment with Aloe vera extract (N=7); and Sepsis + Aloe - Cecal ligation and puncture was performed followed by treatment with Aloe vera extract (N=7). The animals were observed until death. Kidneys were analysed by histopathology. Kruskall-Wallis test was used to compare the histopathological results. Survival analysis was assessed by Kaplan-Meier statistics and compared using log rank test. A significance level of 5% was adopted. The survival time ranged from 19 hours for the Control group up to 24 hours for the animals of Sepsis + Aloe group, corresponding to a survival time of 1.26 times greater (p0.05). Conclusions: Aloe vera extracts delays mortality but does not attenuate kidney injury in sepsis induced by cecal ligation and puncture in mice.

The processed Aloe vera gel (PAG) contains immunomodulatory polysaccharides, which consists of a mixture of polymer chains of b-(1,4)-linked acetylated galactomannan with different lengths. PAG was shown to inhibit azoxymethane(AOM)-initiated and dextran sodium sulfate(DSS)-promoted colon carcinogenesis model in mice. In the present study, we examined the molecular mechanisms for the inhibition of colitis-associated colon carcinogenesis by PAG. PAG administration significantly inhibited the production of pro-inflammatory cytokines and mediators in the colon. Western blot analysis using the colon extracts showed that PAG administration reduced the activation of NF-kB resulting in the inhibition of iNOS and COX-2 expression. PAG appeared to inhibit NF-kB activation through the PPARγ activation. PAG administration also inhibited the expression and phosphorylation of STAT3, which was known to connect inflammation and cancer. PAG administration also inhibited cell cycle progression-related transcription factors such as the proliferating cell nuclear antigen (PCNA), ERK, cyclin-dependent kinase (CDK) 4 and cyclin D1. In contrast, PAG administration increased the expression of CDX2, which was known to be a tumor suppressor in colorectal cancers. These findings show that PAG suppresses colitis-related colon carcinogenesis by inhibiting both chronic inflammation and cell cycle progression in the colon.
