Which is why nutrient deficiencies are common. And not getting an adequate supply of certain nutrients can contribute to poor joint health, weak muscles, and pain. For starters, amino acids are the building blocks of protein. They are used to build muscles as well as collagen, which is the most abundant protein in your body. It’s found in your tendons, ligaments, cartilage, bones, skin, and blood vessels among other places. So if your body isn’t efficiently digesting and absorbing protein, joint and muscle-related problems are possible. Vitamin C is also necessary to build and repair collagen, and a deficit is common among those with gluten sensitivity. Plus, scurvy is a muscle and joint pain disease caused by a vitamin C deficiency. A study found that those with muscle pain are often deficient in selenium and zinc. And it’s suggested that selenium and zinc play a role in both reducing inflammation and preventing tissue damage. Animal studies have found that vitamin B12 may help regenerate nerves as well as prevent pain signaling in the brain. Clinical human trials have also found vitamin B12 to be an effective treatment for lower back pain and neuralgia (nerve-related pain). Omega-3 fatty acids are known to block inflammation. However, they’re not found in grains or processed foods made with them. So deficiencies are common. Calcium is a mineral responsible for muscle contractions, while magnesium is a mineral necessary for relaxing your muscles. Thus, magnesium deficiency may cause muscle cramps. In animal studies, magnesium has been shown to block pain signals. And in humans, it’s been shown to help relieve both acute and chronic pain. Finally, vitamin D plays a role in bone, muscle, and joint health. Deficiencies have been associated with inflammatory pain. And treatment with vitamin D shows promise for those with muscular pain when deficiencies are present. The bacillus Calmette-Guérin (BCG) vaccine is one of the oldest vaccines in history and has been investigated as a potential treatment for type 1 diabetes by Dr. Denise Faustman and her team at the Massachusetts General Hospital (MGH) for over a decade. A new study published today in npj Vaccines describes the long-term outcomes of BCG administration to patients with type 1 diabetes. The study enrolled 282 individuals, 52 of whom received the vaccine. The average age of type 1 diabetes duration among the cohort was 19 years. The patients were followed for over eight years, and their A1c levels were evaluated. The study reports that patients with type 1 diabetes who received just two doses of the BCG vaccine, spaced four weeks apart, achieved markedly lower A1c levels compared to the control group. Nutrients Free Full-Text Effect of Trehalose Supplementation on Autophagy and Cystogenesis in a Mouse Model of Polycystic Kidney Disease
Autophagy impairment has been demonstrated in the pathogenesis of autosomal dominant polycystic kidney disease (ADPKD) and could be a new target of treatment. Trehalose is a natural, nonreducing disaccharide that has been shown to enhance autophagy. Therefore, we investigated whether trehalose treatment reduces renal cyst formation in a Pkd1-hypomorphic mouse model. Pkd1 miRNA transgenic (Pkd1 miR Tg) mice and wild-type littermates were given drinking water supplemented with 2% trehalose from postnatal day 35 to postnatal day 91. The control groups received pure water or 2% sucrose for the control of hyperosmolarity. The effect on kidney weights, cystic indices, renal function, cell proliferation, and autophagic activities was determined. We found that Pkd1 miR Tg mice had a significantly lower renal mRNA expression of autophagy-related genes, including atg5, atg12, ulk1, beclin1, and p62, compared with wild-type control mice. Furthermore, immunohistochemical analysis showed that cystic lining cells had strong positive staining for the p62 protein, indicating impaired degradation of the protein by the autophagy-lysosome pathway. However, trehalose treatment did not improve reduced autophagy activities, nor did it reduce relative kidney weights, plasma blood urea nitrogen levels, or cystatin C levels in Pkd1 miR Tg mice. Histomorphological analysis revealed no significant differences in the renal cyst index, fibrosis score, or proliferative score among trehalose-, sucrose-, and water-treated groups. Our results demonstrate that adding trehalose to drinking water does not modulate autophagy activities and renal cystogenesis in Pkd1-deficient mice, suggesting that an oral supplement of trehalose may not affect the progression of ADPKD.
