Tubulointerstitial fibrosis is recognized as a key determinant of progressive chronic kidney disease (CKD). Fucoidan, a sulphated polysaccharide extracted from brown seaweed, exerts beneficial effects in some nephropathy models. The present study evaluated the inhibitory effect of oligo-fucoidan (800 Da) on renal tubulointerstitial fibrosis. We established a mouse CKD model by right nephrectomy with transient ischemic injury to the left kidney. Six weeks after the surgery, we fed the CKD mice oligo-fucoidan at 10, 20, and 100 mg/kg/d for 6 weeks and found that the oligo-fucoidan doses less than 100 mg/kg/d improved renal function and reduced renal tubulointerstitial fibrosis in CKD mice. Oligo-fucoidan also inhibited pressure-induced fibrotic responses and the expression of CD44, β-catenin, and TGF-β in rat renal tubular cells (NRK-52E). CD44 knockdown downregulated the expression of β-catenin and TGF-β in pressure-treated cells. Additional ligands for CD44 reduced the anti-fibrotic effect of oligo-fucoidan in NRK-52E cells. These data suggest that oligo-fucoidan at the particular dose prevents renal tubulointerstitial fibrosis in a CKD model. The anti-fibrotic effect of oligo-fucoidan may result from interfering with the interaction between CD44 and its extracellular ligands.

Physical Stress Induced Reduction of Proliferating Cells and Differentiated Neuroblasts Is Ameliorated by Fermented Laminaria japonica Extract Treatment
Laminaria japonica is widely cultivated in East Asia, including South Korea. Fucoidan, a main component of L. japonica, protects neurons from neurological disorders such as ischemia and traumatic brain injury. In the present study, we examined the effects of extract from fermented L. japonica on the reduction of proliferating cells and neuroblasts in mice that were physically (with electric food shock) or psychologically (with visual, auditory and olfactory sensation) stressed with the help of a communication box. Vehicle (distilled water) or fermented L. japonica extract (50 mg/kg) were orally administered to the mice once a day for 21 days. On the 19th day of the treatment, physical and psychological stress was induced by foot shock using a communication box and thereafter for three days. Plasma corticosterone levels were significantly increased after exposure to physical stress and decreased Ki67 positive proliferating cells and doublecortin immunoreactive neuroblasts. In addition, western blot analysis demonstrated that physical stress as well as psychological stress decreased the expression levels of brain-derived neurotrophic factor (BDNF) and the number of phosphorylated cAMP response element binding protein (pCREB) positive nuclei in the dentate gyrus. Fermentation of L. japonica extract significantly increased the contents of reduced sugar and phenolic compounds. Supplementation with fermented L. japonica extract significantly ameliorated the increases of plasma corticosterone revels and decline in the proliferating cells, neuroblasts, and expression of BDNF and pCREB in the physically stressed mice. These results indicate that fermented L. japonica extract has positive effects in ameliorating the physical stress induced reduction in neurogenesis by modulating BDNF and pCREB expression in the dentate gyrus.

This work determines the effect nopal consumption at different maturity stages (60, 200, 400, and 600 g) as the only calcium source in bone metabolism. The apparent mineral absorption, the biomarkers of bone metabolism, the bone mineral density at different femoral regions, and crystal properties of the bone were evaluated during the growth stage. The Ca absorption was increased with the rat age in most of the experimental groups, while Mg supplementation decreased intestinal absorption probably due to a saturation process. Intestinal Ca and Mg absorption showed an opposite trend; this result suggests that both ions can compete for vitamin D absorption sites. The percentage of absorption of K was lower in the groups fed with Nopal; nevertheless, due to supplementation, the net absorption was higher than the control group. In all groups, osteocalcin levels decreased with the rat age. Nopal consumption increased osteocalcin levels during the adolescence stage in comparison to the control group. Amino N-terminal propeptide of type I procollagen levels increased in puberty and adolescence in all groups compared to the control group. Bone mineral density in different femoral regions was lower in the groups fed with nopal at early maturity stages (N-60 and N-200) than the groups fed with nopal at late maturity stages (N-400 and N-600). The crystal size of hydroxyapatite exhibited changes for all the groups, indicating the inclusion of mono and divalent ions in calcium replacement. On this basis, the nopal at late maturity stage contributed to bone formation.

Soluble and insoluble fractions from nopal (Opuntia ficus) cladodes improve postprandial glycaemic regulation a crossover randomized study in healthy volunteers
Background and objectives: Type 2 diabetes has been increasing all over the world. A healthy diet, including particular functional foods is an effective strategy to prevent this form of diabetes. Nopal (Oputia ficus) is a cactus plant that, according to traditional medicine, has antidiabetic properties. The objective of this research was to examine the effects of different fractions of nopal cladodes on postprandial glycaemic regulation in healthy volunteers. Methods: Nopal cladodes flour was separated by centrifugation into two fractions (a water insoluble and a soluble fraction). The fractions were incorporated into two different test breads, i.e. insoluble fraction bread (INB) and soluble fraction bread (SNB), and evaluated with respect to acute glycaemic properties in 17 healthy subjects with normal body mass index, applying a randomized cross-over study design. The effects on glycaemic and insulinaemic responses were compared with the responses after a control product, which composed of a similar bread lacking the nopal fractions (white wheat bread, WWB). The products were characterized with respect to starch and dietary fibre content. In addition, starch hydrolysis index (HI) was determined with an in-vitro method in order to predict glycaemic index (pGI),and elucidate possibly underlying mechanisms on postprandial glycaemia. Results: Results regarding glycaemic regulation show that insoluble nopal fractions had a beneficial effect on postprandial glucose regulation. Consequently, the incremental area under the postprandial blood glucose curve (iAUC) after INB was significantly reduced compared to after intake of the WWB during the time periods 0 – 45 and 0 – 60 minutes (p < 0.05). In addition, the iPeak value, i.e. the individual highest incremental peak concentrations, was significantly lowered after INB compared to WWB (P < 0.05). Regarding the postprandial insulin secretion, both SNB and INB lowered the postprandial iAUC compared to the WWB during the time period 0 – 45 and 0 – 60 minutes (P < 0.05). On the other hand, only the INB significantly reduced the insulin iPeak value compared to WWB (P < 0.05). In terms of HI, INB had lower HI and pGI compared to WWB and SNB. Also, INB exhibited highest total, soluble and insoluble dietary fibre contents. Conclusion: The INB promoted lower postprandial glucose and insulin response, while SNB showed beneficial effects only on postprandial insulin secretion. The beneficial effects of nopal on glycaemic regulation might be to some extent explained by the dietary fibre content, especially the INB. However, with respect to the improved postprandial insulin economy observed after the SNB, additional mechanisms are probably involved.

Insulin entrapped alginate-gum tragacanth (ALG-GT) hydrogels at different ALG replacement ratios (100, 75, 50, 25) were prepared through an ionotropic gelation method, followed by chitosan (CH) polyelectrolyte complexation. A mild gelation process without the use of harsh chemicals was proposed to improve insulin efficiency. Retention of almost the full amount of entrapped insulin in a simulated gastric environment and sustained insulin release in simulated intestinal buffer indicated the pH sensitivity of the gels. Insulin release from hydrogels with different formulations showed significant differences (p < 0.05). Time domain (TD) NMR relaxometry experiments also showed the differences for different formulations, and the presence of CH revealed that ALG-GT gel formulation could be used as an oral insulin carrier at optimum concentrations. The hydrogels formulated from biodegradable, biocompatible, and nontoxic natural polymers were seen as promising devices for potential oral insulin delivery.

The results were determined using enzyme-linked immunosorbent assay, quantitative real-time polymerase chain reaction, Western blotting, and immunohistochemical staining. Fucoidan from F. vesiculosus could inhibit the activation of hepatic stellate cells and the formation of extracellular matrix and autophagosomes, and its effect may be associated with the downregulation of transforming growth factor beta 1/Smads pathways. Fucoidan, as an autophagy and transforming growth factor beta 1 inhibitor, could be a promising potential therapeutic agent for liver fibrosis. Background Aspirin is one of the most popular NSAIDs worldwide because of its anti-inflammatory and anticoagulant effects, and however, gastrointestinal injury remains a major complication. We previously reported co-lyophilized aspirin/trehalose (Lyo A/T) decreased the aspirin-induced gastric lesions in dogs. Aim This study investigated the mechanism of gastroprotective effects of trehalose in vitro and in vivo. Methods The apoptotic assays were performed in a human gastric carcinoma cell line, which was treated with aspirin, mixed aspirin/trehalose (Mix A/T) or Lyo A/T. Gastric ulcer severity was examined after oral administration of drugs in rats. In addition, the mucosal tissue apoptotic status in drug-treated rats was evaluated. Molecular dynamics simulations and laser Raman spectroscopy were performed in order to examine the molecular properties of Lyo A/T. Results DNA fragmentation was detected in AGS cells that were treated with aspirin and Mix A/T, but not in the Lyo A/T-treated cells. There were fewer apoptotic cells in the Lyo A/T-treated cells than in the other cells. Gastric injury was reduced in rats that received oral Lyo A/T compared with the others, while PGE2 synthesis was equally decreased in all groups. TUNEL assay and immunohistochemistry of cleaved caspase-3 in the mucosal tissues also revealed that Lyo A/T treatment induced less apoptosis than the others. The Lyo A/T spectrum showed clear differences in several Raman bands compared with that of Mix A/T. Conclusions Our data showed that co-lyophilization of aspirin with trehalose reduced gastric injury, potentially through suppression of aspirin-induced mucosal cell apoptosis while retaining its anti-inflammatory effects.

Fucoidan is a unique bioactive and dietary polymer enriched mainly in the cell wall matrix of the brown seaweeds. This present study was intended to reveal the antigenotoxicity effect of fucoidan on 4-nitroquinolin-1-oxide (4-NQO) induced genetics damage and apoptosis in mice bone marrow cells. The 4-NQO caused genetic damages in the form of chromosome/chromatic breakage was estimated by micronuclei assay whereas apoptosis by annexin-V FITC kit and DNA damage by comet assay kit. In addition, oxidative damage in terms of plasma lipid peroxidation (LPO) and 8-OHdG was also estimated. In the experimental regime, six groups with each in five either sex of mice were used. Fucoidan constituted (50,100,200 mg/kg bwt) by orally for 5 days consequently and on 6th day, 4-NQO was administered (7.5 mg/kg bwt) by i.p. The results clearly show that negative control (H2O) and fucoidan alone constituted mice were not exhibited significant effect on LPO, genetic damages whereas positive control group (4-NQO 7.5 mg/kg bwt, i.p.) showed significant effect on genetic damage by showing increased level of LPO (6.25 vs 1.3 μM MDA), 8-OHdG (12 vs 4%), micronuclei about six-fold, 5-fold of comet, and 4-fold of apoptosis when compared with negative control, 11.6 ± 2.07, 5.00 ± 1.58, and 4.14 ± 0.65 respectively. Fucoidan pretreatment significantly protected the 4-NQO-induced genetic damage by 77% decreased level of micronuclei and 96% comet at dose of 200 mg/kg bwt over the positive control whereas LPO, 8-OHdG, and apoptosis were restored as equal to negative control. This study found as fucoidan possessing significant antigenotoxicity property by protecting 4-NQO-induced genetic damage in mice bone marrow cells as dose dependent manner suggest as valuable food supplements and medicine for mankind from environmental toxicants.

In Vitro Immunomodulatory Effect of Food Supplement from Aloe vera
Food industries typically use Aloe vera as concentrated (100&#xd7; to 200&#xd7;) and dried powders in their final products. These powders are obtained by extrusion of Aloe inner leaf gel (ILG) or Aloe whole leaf (WLP); the juice is filtered through diatomaceous earth and activated carbon before spray drying at temperatures below 70 &#xb0;C. In another process, Aloe inner leaf gel was dried at &#x7e;80 &#xb0;C and mashed to a powder rich in high molecular weight fibres and soluble polysaccharides (ILF). In contrast to ILG and WLP, the ILF sample was cytotoxic for the human intestinal cell line Caco-2 (CC50 = 1&#x2009;g/l), even at concentrations below the recommended dose for human consumption. At lower concentrations (250&#x2009;mg/l) with LPS challenged macrophage-like THP-1 cells decreased by 40&#x25; the release of the anti-inflammatory cytokine IL-10, whereas the release of the proinflammatory cytokine IL-1&#x3b2; increased by 35&#x25; (compared to untreated but challenged macrophage-like THP-1 cells). Unexpectedly, under the same conditions, the less cytotoxic ILG and WLP, both samples with a lower fibre content, significantly increased (up to 2.4 times) the release of IL-10, while the concentration of IL-1&#x3b2; remained unaltered and of TNF&#x3b1; decreased by 35&#x25;. Even more interesting is that a treatment of the ILF sample with activated carbon reduced its cytotoxicity and increased the IL-10 release (3.1 times). Based on these results, we suggest applying an activated carbon treatment on Aloe-starting products, which have high fibre content and have received high temperature treatment, in order to reduce their cytotoxicity and improve their immunomodulatory properties.
