Probiotic Supplementation in a Clostridium difficile-Infected Gastrointestinal Model Is Associated with Restoring Metabolic Function of Microbiota
Clostridium (C.) difficile-infection (CDI), a nosocomial gastrointestinal disorder, is of growing concern due to its rapid rise in recent years. Antibiotic therapy of CDI is associated with disrupted metabolic function and altered gut microbiota. The use of probiotics as an adjunct is being studied extensively due to their potential to modulate metabolic functions and the gut microbiota. In the present study, we assessed the ability of several single strain probiotics and a probiotic mixture to change the metabolic functions of normal and C. difficile-infected fecal samples. The production of short-chain fatty acids (SCFAs), hydrogen sulfide (H2S), and ammonia was measured, and changes in microbial composition were assessed by 16S rRNA gene amplicon sequencing. The C. difficile-infection in fecal samples resulted in a significant decrease (p < 0.05) in SCFA and H2S production, with a lower microbial alpha diversity. All probiotic treatments were associated with significantly increased (p < 0.05) levels of SCFAs and restored H2S levels. Probiotics showed no effect on microbial composition of either normal or C. difficile-infected fecal samples. These findings indicate that probiotics may be useful to improve the metabolic dysregulation associated with C. difficile infection.

ehalose on its underlying mechanisms in animal models of selected neurodegenerative disorders (tau pathology, synucleinopathy, polyglutamine tract, and motor neuron diseases). All animal studies on neurodegenerative diseases treated with trehalose published in Medline (accessed via EBSCOhost) and Scopus were considered. Of the 2259 studies screened, 29 met the eligibility criteria. According to the SYstematic Review Center for Laboratory Animal Experiment (SYRCLE) risk of bias tool, we reported 22 out of 29 studies with a high risk of bias. The present findings support the purported role of trehalose in autophagic flux and protein refolding. This review identified several other lesser-known pathways, including modifying amyloid precursor protein processing, inhibition of reactive gliosis, the integrity of the blood-brain barrier, activation of growth factors, upregulation of the downstream antioxidant signaling pathway, and protection against mitochondrial defects. The absence of adverse events and improvements in the outcome parameters were observed in some studies, which supports the transition to human clinical trials. It is possible to conclude that trehalose exerts its neuroprotective effects through both direct and indirect pathways. However, heterogeneous methodologies and outcome measures across the studies rendered it impossible to derive a definitive conclusion. Translational studies on trehalose would need to clarify three important questions: 1) bioavailability with oral administration, 2) optimal time window to confer neuroprotective benefits, and 3) optimal dosage to confer neuroprotection....

Disaccharide glasses are increasingly used to immobilize proteins at room temperature for structural/functional studies and long-term preservation. To unravel the molecular basis of protein immobilization, we studied the effect of sugar/protein concentration ratios in trehalose or sucrose matrixes, in which the bacterial photosynthetic reaction center (RC) was embedded as a model protein. The structural, dynamical, and H-bonding characteristics of the sugar–protein systems were probed by high-field W-band EPR of a matrix-dissolved nitroxide radical. We discovered that RC immobilization and thermal stabilization, being independent of the protein concentration in trehalose, occur in sucrose only at sufficiently low sugar/protein ratios. EPR reveals that only under such conditions does sucrose form a microscopically homogeneous matrix that immobilizes, via H-bonds, the nitroxide probe. We conclude that the protein immobilization capability depends critically on the propensity of the glass-forming sugar to create intermolecular H-bond networks, thus establishing long-range, homogeneous connectivity within the matrix.

PURPOSE: To compare the therapeutic effect of sodium hyaluronate (SH)–trehalose (Trehalube, Microlabs, Bangalore, India, SH 0.1% and trehalose 3%) or SH (0.1% Hylotears, Raymed, Chandigarh, India) alone in patients with dry eye disease (DED).MATERIALS AND METHODS: Patients were randomized into two groups: SH-trehalose (SH 0.1% and trehalose 3%) or SH (0.1% Hylotears) alone. The Ocular Surface Disease Index (OSDI) questionnaire was used to assess patient's symptoms. Patients were followed up at 4 and 8 weeks, and OSDI score, tear film break-up time (TBUT), tear film height (TFH), Schirmer's test, and conjunctival staining were evaluated at each visit.RESULTS: A total of 384 patients were included in the study, 192 patients in each arm. The mean age of participants was 37.62 + 14.4 years and 225 were women (56%). The improvement in Schirmer's test was significantly better in the SH-trehalose group at 8 weeks (5.26 + 4.3 mm, 95% confidence interval = 4.6–5.9 mm) compared to the SH group (3.71 + 3.9, 95% confidence interval = 3.15–4.28 mm). The TBUT and TFH showed slight improvement at 4 weeks in both groups, but not at 8 weeks. There were no group differences at all-time points in terms of conjunctival staining and OSDI-based grades of DED.CONCLUSION: It was found that treating dry eye with SH-trehalose leads to greater improvement in the Schirmer's values and TBUT after 8 weeks of sustained use in patients with DED, and this was more pronounced in those with severe DED.

Background For scavenging reactive oxygen species, plant possess effective system that protect them from destructive oxidative reaction. Parts of this system as osmoprotectants and antioxidative enzymes are key elements in the defense mechanisms. A field experiment was conducted to evaluate the potential of foliar treatment of trehalose (Tre) with different concentrations (0, 0.1 mM, or 0.5 mM) in improving antioxidant defense system of quinoa plant under normal irrigation and drought stress conditions. Results Drought stress caused significant increases in some osmoprotectants as glucose, trehalose, TSS, free amino acids, and proline. Meanwhile, trehalose foliar treatment with different concentrations significantly decreases in free amino acids and proline contents. More accumulation of the tested organic solutes of leaves (glucose, sucrose, trehalose, TSS) of the trehalose-treated plant in both normal irrigated and drought-stressed quinoa plants as compared with the corresponding controls. Treating quinoa plants with trehalose resulted in significant decrease in lipid peroxidation, hydrogen peroxide contents, and LOX activity in normal irrigated and drought-stressed plants. These decreases correlated with significant increases in total phenolic contents as compared with untreated control. Different concentrations of trehalose resulted in significant increases in antioxidant enzymes. Maximum increase antioxidant enzymes were observed by treating plants Tre at 0.5 mM either under normal irrigation or drought conditions. Conclusion It could be concluded that foliar spray of trehalose was effective in improving quinoa performance by reducing hydrogen peroxide free radical and by enhancing antioxidant compounds (phenolics), compatible osmolytes, membrane stability, and antioxidant enzymes.

Insulin is an important therapeutic protein for the treatment of diabetes, but it is unstable and aggregates upon exposure to environmental stressors encountered during storage and transport. To prevent degradation of the protein in this manner and retain as much in vivo bioactivity as possible, a well-defined insulin–trehalose glycopolymer conjugate was synthesized. To accomplish this, a strategy was employed to site-specifically modify insulin with a polymerization initiator at a particular conjugation site; this also facilitated purification and characterization. Lysine of the B chain was preferentially modified by conducting the reaction at high pH, taking advantage of its higher nucleophilicity than the N-terminal amines. Trehalose monomer was polymerized directly from this macroinitiator to form a well-defined conjugate. Bioactivity of the site-specific conjugate was shown to be higher compared to the nonspecific conjugate and the same as the analogous site-specific poly(ethylene glycol) (PEG) conjugate, as confirmed by the insulin tolerance test (ITT) in mice. The conjugated trehalose glycopolymer also stabilized insulin to heat as measured by high-performance liquid chromatography (HPLC).

The Effect of Trehalose in Inhibiting Liver Damage via The Il-6 Pathway in Old Mice International Journal Papier Advance and Scientific Review
Trehalose is a non-reducing disaccharide consisting of two glucose molecules linked by a 1–1 glycosidic bond. Trehalose can play a role in organ detoxification, antioxidant enzyme activity, reducing lipid peroxidation and reducing the secretion of inflammatory factors TNF-, IL-1β, Il-6, thereby inhibiting liver damage. Because of its role, this study aims to determine the effect of trehalose administration on liver damage through the IL-6 expression pathway with experimental post-test design with control group design. This research was conducted using samples of old rats, namely Wistar rats (Rattus novergicus). The old rats used were 21 male species which were then divided into 3 groups, namely the old control group, the old rat group that was given sucrose, and the old rat group that was given trehalose. Then observed for 8 weeks, namely in March - May 2021 at the Faculty of Medicine, Hasanuddin University Makassar. The results obtained from the 3 groups showed a significant value that the administration of trehalose sugar in elderly mice had an effect on reducing the inflammatory factor IL-6 because it activated autophagy in macrophages thereby reducing cytokine production and vascular inflammation so as to be able to restore all aging mice to a level like observed in young mice. It is also known that aged rat liver, which is characterized by ER UPR activation and inflammatory signaling, decreases nicotinamide and UDP-N-acetyl-hexosamines, and reduces proteasome activity. Therefore, trehalose can be an effective therapeutic strategy against age-related disorders of proteostasis in the liver.
